赤眼鳟TRIF基因cDNA克隆及其表达特性研究

摘要
本文采用RACE术获得了赤眼鳟(Squaliobarbus curriculus)TRIF(TIR-domain-containing molecule 1)基因的cDNA全长,命名为ScTRIF,ScTRIF基因cDNA序列全长2263bp,开放阅读框(ORF)1677 bp,共编码559个氨基酸,推导的蛋白分子量约为62.70 kDa,理论等电点p I为4.91,SMART结构域测显示,翻译的氨基酸序列具有潜在TIR结构域。赤眼鳟TRIF的氨基酸与草鱼的同源性最高(93%),Real-time PCR结果显示:TRIF在赤眼鳟12个组织中均有表达,其中鳃中的相对表达量最高,脑组织中的表达量最低;在草鱼呼肠弧病毒(Grass carp reovirus,GCRV)感染后,ScTRIF在头肾和脾脏组织中的表达量出现两个上调波动周期,分别在12h和96h上调至峰值。研究表明ScTRIF参与赤眼鳟抗GCRV病毒的免疫应答反应。
The full cDNA sequence of TIR-domain-containing molecule 1(TRIF) in the barbel chub Squaliobarbus curriculus, designated as ScTRIF, has been cloned by using RACE-PCRs method.The full-length of ScTRIF cDNA was 2263 bp, which contained 1677 bp ORF encoding599 amino acids. The theoretical molecular weight and isoelectric point(PI) of ScTRIF were62.7004 k D and 4.91, respectively. The ScTRIF protein possess a TIR domain. According to the comparison with known TRIF amino acid sequences, the putative protein of ScTRIF showed the highest identities with Ctenopharyngodon idellus(93%). The expression of TRIF in different tissues were also detected by Real-time PCR The highest expression level were observed in the gill and the lowest expression level was found in brain. After injection with GCRV, the ScTRIF expression level was obviously down-regulated in the immune tissue at 6h and Sharp increases in12 h and 96 h, respectively. two groups all showed a trend of fluctuation., ScTRIF might play important roles during the immune response of barbel chub against GCRV.
