应用解淀粉芽孢杆菌发酵黄芪培养基的筛选与优化

摘要
筛选优化解淀粉芽孢杆菌固态发酵黄芪的培养基,以菌生长量为指标,采用单因子试验对菌株适宜发酵培养基成分进行筛选;采用二次正交旋转组合试验设计及响应面法,优化了黄芪固态发酵培养基。结果表明,最适发酵培养基成分为黄芪、黄豆粉和CaCO_3。用其分别替代基础培养基中相应营养物质,然后接种解淀粉芽孢杆菌,置37℃以150 r/min振荡培养24h,细菌菌数分别为6.50×10~9 CFU/mL、5.17×10~9 CFU/mL和5.07×10~9 CFU/mL。优化的固态发酵培养基为:黄芪粉30%、黄豆粉10%、CaCO_3 0.2%、水59.8%,37℃发酵培养72 h,发酵物中活菌数达7.67×10~8 CFU/g。本研究优化的黄芪固态发酵培养基适宜解淀粉芽孢杆菌生长,筛选培养基原材料符合固态发酵产业化生产条件。
In order to screening medium for solid fermenting Radix Astragalus by B.amyloliquefaciens,the effective components of solid fermentation medium was selected by the single factor experiments utilising the viable count as an indicator.The components of solid fermentation medium was optimised by quadratic orthogonal rotation combination design and response surface methodology.The results that the optimum components of medium are Astragalus,Soybean flour and Calcium carbonate.The corresponding nutrients in basal medium was replaced by optimum components,then inoculated culture bacteria under the fermentation conditions at temperature 37℃,150 r/min for 24 h,the viable count of B.amyloliquefaciens were 6.50×10~9 CFU/mL,5.17×10~9 CFU/mL and 5.07×10~9 CFU/mL,respectively.The optimum medium consist of 30%Astragalus,10%Soybean flour,0.2%Calcium carbonate and 59.8%water.The viable count of B.amyloliquefaciens cultured in the optimised medium for 72 h at 37℃ was 7.67×10~8 CFU/g.The optimised solid fermentation medium was suitable for the growth of B.amyloliquefaciens,materials are ideal for industrial production conditions of solid fermentation.
