富硒滑子蘑菌丝体和子实体中谷胱甘肽及其合成关键酶的研究

摘要
谷胱甘肽做为体内一种重要的抗氧化剂,能够清除掉人体内的自由基,提高人体免疫力。本论文研究了不同富硒浓度对滑子蘑的菌丝体和子实体中谷胱甘肽及其合成关键酶的影响,培养浓度为:0、20、40、60、80、100mg/kg。采用南京建成生物工程研究所购买的γ-谷氨酰半胱氨酸合成酶(γ-GCS)测定试剂盒测定酶活,四氧嘧啶法测定谷胱甘肽的含量。实验结果:菌丝体和子实体中的γ-GCS活性及谷胱甘肽含量随着富硒浓度则增加呈下降趋势但均大于相对应的空白组;子实体中的γ-GCS活性大于菌丝体中的γ-GCS活性,富硒浓度为20 mg/kg时,菌丝体及子实体的γ-GCS具有最大活性,分别是0.67U/g、1.80 U/g;菌丝体中的谷胱甘肽含量基本上和子实体中的谷胱甘肽含量相同且都高于相对应的空白组,在富硒浓度为20 mg/kg时,菌丝体及子实体的谷胱甘肽含量达到最大值,分别为308.0mg/g、278.0mg/g。进而得到结论,富硒可以促进滑子蘑谷胱甘肽的合成,低浓度富硒的促进效果要优于高浓度富硒。
Glutathione as an important antioxidant in vivo,can rid the body of free radicals,enhance human immunity.This paper studied the effect of different concentration of Se-enriched Pholita Namek mycelium and fruiting body of its glutathione and key synthase,the training concentration as:0,20,40,60,80,100 mg/kg.Using the γ-glutamyl cysteine synthetase(γ-GCS) activity assay kit that purchased from Nanjing Jiancheng Bioengineering Institute to detect activity,the content of glutathione was measured by alloxan method.The results:γ-GCS activity and the content of glutathione in mycelium and fruiting body was in downward trend overall with the increasing concentration of selenium and each of them were greater than the corresponding blank group;γ-GCS activity in fruiting body was higher than that of mycelium,in the Se-enriched concentration of 20 mg/kg,the γ-GCS activity in mycelium and fruiting body having maximum value,namely 0.67 U/g,1.80 U/g;Glutathione content in fruiting body and mycelium were generally equal and both of them were larger than the corresponding blank group,in the Se-enriched concentration of 20 mg/kg,the glutathione content in mycelium and fruiting body reached maximum value,308.0 mg/g,278.0 mg/g.Further conclude that Se-enriched could promote the synthesis of glutathione in Pholita Namek,and low concentration of Se-enriched promoting effect is superior to the high concentration.
