阳离子脂质体的环境响应性评价

摘要
随着人们在细胞分子水平上对免疫缺陷病、肿瘤等多种疾病发病机制认识的不断深入,基因治疗已日渐成为科学家们研究的热点。在基因治疗的基础研究和临床试验中,阳离子脂质体是一类应用最为广泛的非病毒基因载体之一。近年来研究表明,具有pH敏感特性的阳离子脂质体能够对周围环境pH变化进行响应,通过结构自身适应改变实现对基因的控制释放,因此在基因转运研究中备受关注。本研究以课题组合成的阳离子端基肽类脂制备的脂质体作为载体,以不同孵育时间改变该脂质体pH值,通过琼脂糖凝胶电泳实验考察孵育时间和pH值对脂质体与DNA结合能力的影响,研究脂质体的p H敏感响应性能。实验结果表明,改变p H值,阳离子脂质体与DNA结合能力随之改变,在酸性条件下脂质体对DNA的结合能力下降。本研究通过一系列实验,获得脂质体/DNA复合物的最佳pH敏感值,为合成新型环境响应性pH敏感阳离子脂质体做铺垫,从而有望获得最佳的基因治疗效果。
In the research and clinical application of gene therapy, cationic liposome is one of the most important non-viral gene vectors. Recent studies showed that pH sensitive cationic liposomes can respond to environmental changes in pH by changing the structure itself to adapt to achieve controlled release of the gene, which lead to better gene transfer effect. In this study, the acid sensitive response of cationic liposome was evaluated by agarose gel electrophoresis. The experimental results showed that changing the p H, binding of DNA and cationic liposomes changed, under acidic conditions liposome DNA binding capacity decreased. Through a series of experiments, we got the best value of pH sensitive liposome/DNA complexes for the synthesis of novel environmental responsive p H sensitive cationic liposomes pave the way, which is expected to get the best effect of gene therapy.
