波纹唇鱼芳香化酶基因启动子克隆及活性分析

摘要
在硬骨鱼当中,存在性腺型Cyp19a1a和脑型Cyp19a1b两个芳香化酶基因。Cyp19a1a与性别调控及性腺发育直接相关;Cyp19a1b可能间接影响性别分化。本研究在实验室前期所获得的波纹唇鱼Cyp19a1a与Cyp19a1b基因cDNA的基础上采用Genomewalking技术克隆基因的启动子。采用双荧光素酶报告基因检测系统检测启动子活性。在Cyp19a1b启动子上预测到Cp G甲基化结合位点及雌激素受体(ER),而Cyp19a1a启动子都不具备。启动子活性初步分析表明,不同浓度的雄激素能够使Cyp19a1a启动子表达活性提高,而Cyp19a1b则无明显变化。不同浓度的雌激素条件下,Cyp19a1b启动子活性存在显著差异,这与其含有ER有关,而cyp19a1a则无明显变化。本研究为进一步揭示波纹唇鱼芳香化酶基因表达的调控机制奠定基础。
There are two aromatase genes in teleost fish: gonads type Cyp19a1 a and brain type Cyp19a1 b. The level of Cyp19a1 a is directly related to sex control and gonad development, while Cyp19a1 b may affect sexual differentiation indirectly. Based on the findingsof Cheilinus undulatus' cDNA we have got in our previous study, we get the promotersby Genome walking technology. Dual Luciferase Reporter Gene Assay System is used to detect the recombinant plasmid. It is predicted that the Cp G methylation binding sites and estrogen receptor(ER) can be founded in Cyp19a1 b promoter, while in Cyp19a1 a can not. It is showed that different density of androgen could increase the expression of Cyp19a1 a promoter, while Cyp19a1 b has no significant change. Under different density of estrogen, the promoter activity of Cyp19a1 b is significantly different, which bear a close relationship with ER, but no significant relationship with cyp19a1 a.It is hoped that this study can help to clarify the expression and regulation mechanism of P450 and provide a theoretical basis and techniques of artificial reproduction of Cheilinus undulatus.
