中子管中子对小鼠宫颈癌肺转移治疗作用的初步研究

摘要
目的:探讨不同剂量快中子对肺转移模型小鼠的作用及其对荷瘤鼠生存期的影响,为中子治疗肿瘤的临床应用提供实验依据。方法:应用小鼠宫颈癌U14细胞株尾静脉注射法构建小鼠宫颈癌体内肺转移模型;采用不同剂量的快中子对荷瘤鼠全身照射,于照射后第10 d各组随机处死5只实验鼠,HE染色检测肺转移结节大小与数量;其余荷瘤鼠继续常规饲养,直至荷瘤鼠全部死亡,Kaplan-Meier统计方法进行生存期分析。结果:解剖学检测与HE染色均证实U14肺转移模型制作成功。与对照组比较,快中子照射30 min和60 min实验组小鼠肺转移癌结节数减少,荷瘤鼠的生存期延长,90 min实验组小鼠的肺转移癌结节数更少但荷瘤鼠的生存期缩短。结论:适当选择快中子照射剂量可有效抑制癌细胞肺转移,提高荷瘤鼠生存期。
Objective To study the biological effective and life-span of fast neutron on tumor-bearing mice to provide the theoretical basis for further clinical practice.Methods The mice models of pulmonary metastasis by murine cervical cancer were established by U14 cells injection viatail vein,and then the models were divided into four groups at random.On 10 th day after fast neutron radiotherapy,5 mice in each group were sacrificed for the evaluation of tumor inhibition efficacy and the other pulmonary metastasis models were raised and the death time of each model was recorded for Kaplan-Meier survival analysis.Results The mice models were successfully established which were confirmed by anatomy and HE stain assay.The results indicated that the fast neutron inhibited the growth and pulmonary metastasis of U14 in vivo.The mice models treated by fast neutron for 30 min and 60 min gained longer life-span and less metastatic nodules in lung tissues,whereas the mice in 90 min group gained shorter life-span and the least metastatic nodules,compared with the control group,respectively.Conclusions The pulmonary metastasis of U14 were inhibited by fast neutron in a dose-dependent manner,and a proper dosage should be selected for the radiotherapy of tumorbearing mice.
