血清对甲醛和乙醛诱导细胞毒性的影响

摘要
由于醛类的羰基能与蛋白质发生加和反应,因而血清可能对醛类体外实验具有较大的影响。为了解血清在醛类染毒时的影响,以含10%胎牛血清和不含胎牛血清的RPMI-1640培养基作为甲醛和乙醛染毒液,通过CCK8法(线粒体毒性)和LDH法(细胞膜毒性)分别对这两种醛类在有血清和无血清时的A549细胞毒性进行检测。结果表明,CCK8法检测的甲醛和乙醛在无血清组的IC50分别是有血清组的5和2.8倍。LDH法检测的甲醛和乙醛在无血清组的IC50分别是有血清组的0.67和1.5倍。配对t-test表明,CCK8实验中,血清对甲醛和乙醛诱导的线粒体毒性均具有显著影响(p<0.001);LDH实验中,血清对甲醛诱导的细胞通透性改变具有显著影响(p<0.001),而对乙醛诱导的细胞通透性变化没有显著影响(p=0.425)。因此,和无血清组相比,甲醛和乙醛在有血清组的线粒体毒性更高。同时,甲醛在有血清组的细胞膜通透性改变更小,而乙醛则在有无血清存在时对细胞膜的通透性没有明显的影响。
Because carbonyl of aldehydes can react with protein,thus serum may have an important effect on the experiment in vitro for aldehydes.To investigate this effect,A549 cells were exposed to formaldehyde and acetaldehyde in RPMI-1640 medium with or without 10% fetal bovine serum(FBS),and the toxicity was detected by CCK8 assay(mitochondrial toxicity) and LDH assay(cell membrane toxicity).The results showed that the IC50 of the FBS negative group detected with CCK8 assay was 5 and 2.8 times higher than FBS positive group for formaldehyde and aldehyde,respectively.IC50 of the FBS negative group detected with LDH assay was 0.67 and 1.5 times higher than FBS positive group for formaldehyde and aldehyde,respectively.Paired t-test showed that,in CCK8 assay,serum had a significant effect(p < 0.001) on mitochondrial toxicity induced by formaldehyde and acetaldehyde;and in LDH assay,serum on had a significant effect(p< 0.001) on the cell membrane permeability for formaldehyde,but there was no significant effect(p=0.425) for acetaldehyde.Therefore,Compared with FBS negative group,the toxicity level of mitochondrial is higher for formaldehyde and acetaldehyde in FBS positive groups.The change of cell membrane permeability is litter for formaldehyde in contrast with FBS positive group and there is no significant effect for acetaldehyde between FBS negative groups and FBS positive group.
