南川百合种子萌发及试管鳞茎的培育初探

摘要
南川百合为中国特有植物,本研究对组培条件下种子萌发和试管鳞茎增大的影响因素、以及光照和多效唑对移栽后幼苗生长的影响进行了探究,研究结果表明:①南川百合种子萌发与温度、剥种皮处理、基本培养基大量元素含量等因素相关,南川百合种子最适宜的萌发温度为15~20℃;剥种皮处理的种子具有最高的发芽率和发芽势,分别为90.48%、42.86%;使用1/2MS培养基的种子在第9天开始萌发,并且具有较高的发芽势和发芽率。②试管鳞茎的增大与蔗糖浓度及活性炭含量相关,蔗糖浓度为60g/L以及活性炭含量为1.00g/L的培养基能够显著加快试管鳞茎的增大。③施用0.5mg/L的多效唑能够促进南川百合幼苗叶片数量增加,提高其移栽成活率;长日照对于南川百合的幼苗生长没有影响。该研究对南川百合的繁殖和引种驯化具有一定的实际应用价值。
Lilium rosthornii is an endemic plant to China.It aims to study the tissue culture conditions which influenced seed germination,bulblet increasing and the growth of tissue culture seedling,to establish tube bulb cultivation and breeding technology of preliminary system of L.rosthornii under tissue culture conditions.It indicated that L.rosthornii seeds germination was related to temperature,seed coat,culture medium and many factors;the optimum seeds germination temperature was 15-20℃;peeling the coat of the seeds had the highest germination rate,germination potential 90.48%,42.86%;seeds began to germinate in the 9th day under 1/2 MS culture medium and had a higher germination potential and germination rate.The increase of tube bulb was related to the concentration of sucrose and the concentration of activated carbon.The sucrose concentration of 60g/L and the concentration of activated carbon for 1.00g/L could significantly accelerated the growth of tube bulb.The long day for the growth of L rosthornii was not significant,and the application of 0.5 mg/L paclobutrazol can promote the increasing in the number of L.rosthornii seedling leaves and improvement of the survival rate.
