短链氯化石蜡暴露对大鼠代谢通路的影响

摘要
短链氯化石蜡(Short-chain chlorinated paraffins,SCCPs)是碳链长度为10-13,氯化度在30-70%的氯代正构烷烃。SCCPs具有远距离环境迁移能力、环境持久性、生物蓄积性等POPs特性[1,2],于2009年被斯德哥尔摩公约列为POPs候选物质。本研究基于代谢组学技术在最低可观察有害作用水平(100 mg/kg/day)之下观察了SCCPs连续28天暴露对SD大鼠血液小分子代谢物的影响。基于Metabo Analyst 3.0对不同剂量SCCPs暴露后显著变化的代谢物进行分析。结果表明SCCPs暴露显著干扰了SD大鼠体内的类固醇类激素的生物合成、三羧酸循环、甘油磷脂代谢、α-亚麻酸代谢、半乳糖代谢、二元羧酸代谢、鞘脂类代谢、花生四烯酸代谢、色氨酸代谢、谷胱甘肽代谢、亚油酸代谢、谷氨酰胺谷氨酸代谢、丙氨酸,天冬氨酸和谷氨酸代谢、精氨酸和脯氨酸代谢、嘧啶代谢、糖基磷脂酰肌醇锚合成、胆汁酸合成等代谢通路。总体而言,受SCCPs干扰的代谢通路主要为脂质代谢、糖代谢、氨基酸代谢和三羧酸循环相关通路。
Short-chain chlorinated paraffins(SCCPs) have attracted considerable attention for their characteristic of persistent organic pollutants.In this study,metabolomic approach was used to evaluate the effect of SCCPs(< 100 mg/kg/day) on SD rat after 28 days expoure.Metabolic pathway analysis was conducted based ona web service for metabolomics data analysis(Metabo Analyst 3.0).SCCPs exposure caused significant metabolic disruptions with SD rat,including the steroid hormone biosynthesis,citrate cycle,glycerophospholipid metabolism,α-linolenic acid metabolism,galactose metabolism,sphingolipid metabolism,arachidonic acid metabolism,tryptophan metabolism,glutathione metabolism,linoleic acid metabolism,glutamine and glutamic acid metabolism.alanine,aspartate,and glutamate metabolism arginine and proline metabolism,pyrimidine metabolism,glycosylphosphatidylinositol anchor biosynthesis,primary bile acid biosynthesis.The toxic effects of SCCPs might mainly involve the perturbation of lipid metabolism,glucose metabolism and amino acids metabolism.
