一种应用到活细胞中检测锌离子的苯并噁唑类荧光探针

摘要
以2-(2’-羟苯基)苯并噁唑为母体结构,在3’和5’端引入吸电子基团,合成了一种苯并噁唑类荧光探针[1,2]。其结构通过质谱、红外光谱和核磁共振波谱进行了表征并用光谱法研究了其性能。结果表明:该探针激发波长为320 nm,发射波长为471 nm,对Zn~(2+)表现出良好的选择性和灵敏性。与Zn~(2+)作用后,紫外灯下溶液的颜色由淡蓝色变成亮蓝色,发射波长由471 nm移至451 nm处,且荧光发射强度随着Zn~(2+)浓度的增加不断增强,在1–10μM的浓度范围内,具有良好的线性荧光响应,最低检测线为78 n M,结合常数为1.19×10~3 M~(-1)。此外,该探针应用于活细胞中,能跨过细胞膜实现对Zn~(2+)的检测,其细胞毒性低,有应用到生物领域的潜质。
A 2-(2'-hydroxy phenyl) benzoxazole-based fluorescence probe with introduction of electron-accepting group at 3', 5' position had been synthesized. The structure was characterized by MS, FT-IR, NMR and optical properties were investigated by spectrometry. The results showed that the probe excitation wavelength was 320 nm, emission wavelength 471 nm, and had good selectivity and sensitivity to Zn~(2+). Its interaction with Zn~(2+) caused color change from light blue to bright blue under a UV lamp, and a hypochromatic shift in emission from 471 nm to 451 nm, with the fluorescence intensity increasing linearly with the Zn~(2+) concentration in the range of 1–10 μM. The binding constant(Kd) was calculated to be 1.19×10~3 M~(-1) and detection limit was 78 n M. In addition, the cell membrane penetration of probe had low cytotoxicity, and it can be applied to image Zn~(2+) in living cells, indicating its potential application in biological field.
