DVDMS-Mn脂质体用于双模态影像引导的声动力治疗研究

摘要
本研究主要目的是制备DVDMS-Mn脂质体,研究其成像效果与声动力效果。通过采用薄膜-水化法制备DVDMS-Mn脂质体并表征其特性。利用荧光成像及MRI成像验证其成像效果,并通过CCK8试剂盒检测各处理组细胞活性,验证其声动力效果。结果表明:DVDMS-Mn脂质体粒径为85.15±2.14 nm,PDI值为0.21±0.019,表面电位为-2.77±0.34 m V,包封率为65.56±1.47%,Mn与DVDMS的摩尔比为1.6:1。荧光图片及MRI图片均证实DVDMS-Mn脂质体有较好的成像效果。细胞活性试验表明SDT组的细胞活性明显低于其余三组(P<0.05)。我们的研究揭示DVDMS-Mn脂质体不但具有荧光成像和MRI成像功能,而且声动力治疗效果明显,为诊疗一体化提供新思路。
The aim of this project is to prepare DVDMS-Mn-liposomes and evaluate its fluorescence, MRI imaging and SDT effects. DVDMS-Mn-Liposomes were prepared by thin-film rehydration method, and verified the imaging effect by fluorescence imaging and MRI imaging. The SDT effects of DVDMS-Mn-LPs were verified by CCK8. Our results showed the average particle size, PDI and zeta potential of DVDMS-Mn-liposomes were(85.15 ± 2.14) nm, 0.21 ± 0.019 and(-2.77 ± 0.34) m V, respectively. The drug encapsulation efficiency was 65.56 ± 1.47%. The chelation of Mn with DVDMS occurred at a molar ratio of 1.6:1. The DVDMS-Mn-liposomes has a good fluorescence imaging and MRI imaging effects. CCK8 assays displayed that the cell viabilities of SDT group was significantly lower than the other three groups(P<0.05). In conclusion, the DVDMS-Mn-liposomes is a promising sensitizer for SDT under the guidance of fluorescence imaging and MRI imaging.
