一株源自金针菇样品的单增李斯特菌优势株798-1 WT基因组分析

摘要
单增李斯特菌(Listeria monocytogene)是兼性厌氧的革兰氏阳性食源性致病菌,被WHO列为四大重要食源性致病菌之一。我们在前期研究中获得一株源自金针菇样品的单增李斯特菌798-1WT优势株。本研究主要对其进行全基因组测序并进行全基因组比较分析。单增李斯特菌798-1WT全基因组随机片段化处理后,使用Illumina Genome Analyzer IIx测序平台进行de novo测序。单增李斯特菌798-1 WT基因组大小为3,013,905 bp,G+C含量为37.5%,预测共有3057个开放阅读狂,共编码3057个ORF,其中18条rRNA和65条tRNA操纵子。与单增李斯特菌EGD-e参考菌株基因组比较,共发现9个区域出现变异或者缺失,分别为lmo0066,lmo0067,lmo0252,lmo0253,inlG,lmo0363,lmo0364,lmo1097~lmo1102,lmo1289,lmo2026,cornK prophage,lmo2407~lmo2410,lmo2774,lmo 775,lmo2776。单增李斯特菌798-1WT全基因组比较分析为该优势株的表型特征分析提供了清晰的遗传信息,对食用菌生产防控单增李斯特菌污染具有重要的意义。
Listeria monocytogenes,a facultative Gram-positive foodbome pathogen,has been considered as one of the fourth important foodborne pathogen by WHO.Here,we reported the draft genome sequence of L.monocytogenes 798-1WT which isolated from Flammidina velutipes product in Soudi China.The draft genome of L.monocytogenes was sequenced with a wholegenome shotgun strategy using an Illumina Genome Analyzer IIx instrument,which genome is 3,013,905 bp in size with an average G + C content of 37.5%.A total of 3057 ORFs(open reading frames),18 rRNA fragments and 65 tRNAs were identified.Compared to the L.monocytogenes EGD-e,L.monocytogenes 798-1 WT was distinguished by several genes deletions,such as lmo0066,lmo0067,lmo0252,lmo0253,inlG,lmo0363,Zmo0364,lmo1097~lmol102,Zmol289,Zmo2026,comK prophage,lmo2407 ~ lmo2410,lmo2774,lmo2775,lmo2776.This information will give the opportunity to provide genetic basis for clarifying the characterization of this dominated strain.It is of importance for prevention and controlling the persistence L.monocytogenes in mushroom production.
