山月桂组培苗生根实验

摘要
本实验以山月桂‘奥运圣火'组培苗为材料,研究不同植物生长调节剂和凝固剂对其生根的影响,同时研究不同栽培基质对其移栽成活率的影响,为其将来实现工厂化育苗提供依据。结果表明:①同时添加IBA和2-ip更有助于山月桂‘奥运圣火'组培苗生根,并且珍珠岩比琼脂更适合做生根培养的凝固剂,因此最适合的生根培养基为:1/2 WPM+1.0mg·L~(-1)IBA+0.1mg·L~(-1)2-ip+珍珠岩,生根率87.32%,平均根长为4.39cm;②适合组培苗的移栽基质为珍珠岩、泥炭土(1:1,V/V)的混合基质,成活率为46.67%。
In the research,we took the Kalmia.latifolia‘Olympic Fire' as material to study the effects of different plant growth regulator and coagulant on the rooting of plantlet in vitro,as well as the effects of different substrates on the transplanting of tissue cultured seedling.The results showed that;①adding the IBA and 2-ip simultaneously in the culture medium would induce a definite improvement on the rooting of seedlings,and the perlite was more suited to be the coagulant than the agar in the rooting medium.As a consequent,the best medium for rooting of plantlet was 1/2WPM+1.0mg·L~(-1)IBA+0.1mg·L~(-1) 2-ip+perlite,with the rooting rate of 87.32%and the average root length of 4.39cm;②The most suitable media for transplanting was the mixture of perlite and peat soil with the volume ratio of 1:1,and the survival rate was 46.67%.
