针刺结合美多巴对帕金森小鼠模型多巴胺能神经元及脑线粒体复合物酶的影响

摘要
目的探讨针刺百会和大椎穴对帕金森病(PD)小鼠多巴胺能(DA)神经元的保护作用及可能机制。方法 C57BL/6雄性小鼠随机分为5组:正常组、模型组、针刺组、美多巴组、针刺结合美多巴组。应用腹腔注射1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)建立PD动物模型,造模后动物存活56天。采用免疫组织化学技术检测黑质酪氨酸羟化酶(TH)阳性神经元数目,透射电镜观察其神经元的超微结构,并测定各组小鼠脑线粒体复合物的活性。结果与正常组相比,模型组TH阳性神经元数目较正常组减少(P<0.05),针刺组、针刺结合美多巴组较模型组明显增多(P<0.05)。电镜观察下发现模型组神经元有不同程度的损伤,各治疗组神经元损伤有所减轻,针刺结合美多巴组神经元损伤最轻。与正常组相比,模型组小鼠线粒体复合物活性均显著下降(P<0.05);与模型组相比,针刺组线粒体复合物Ⅱ、Ⅲ活性,针刺结合美多巴组线粒体复合物Ⅰ、Ⅳ活性显著上升(P<0.05)。结论针刺百会、大椎穴可能通过抑制脑内线粒体复合物活性的下降来维持其线粒体的功能,以达到保护多巴胺能神经元活性的目的。
Objective To investigate the mechanisms of acupuncture in preventing and treating Parkinson's disease(PD).Methods Male C57BL/6 mice were randomly divided into five groups:control group(C),model group(M),acupuncture group(A),Medopar group(P),acupuncture mixed with Medopar group(AP).The PD model was induced by MPTP with intraperitoneal injection.Different treatments were taken.All mice were sacrificed at the 56 th day respectively after first injecting with MPTP.The TH+(Tyrosine hydroxylase) dopaminergic neurons in the substantia nigra were counted in every group by immunohistochemistry technique.The ultrastructure of neurons and photography were observed by transmission electron microscope.The activity of mitochondrial complex in the brain of mice was measured respectively.Results The number of TH positive neurons in M was much less than that in C(P<0.05).The numbers of the neurons in A and AP were obviously more than that in M(P<0.05).The dopaminergic neurons in M were found damaged more than that in C,the dopaminergic neurons in A,P,AP were found damaged less than that in M,the dopaminergic neurons in AP had the lightest minor damage.The activities of mitochondrial complex Ⅰ,Ⅱ,Ⅲ and Ⅳ in M were significantly lower(P<0.05)when compared with those in C;the activities of mitochondrial complexⅡ,Ⅲ in A andⅠ,Ⅳ in AP were significantly higher(P<0.05)compared with those in the M.Conclusions Needling Baihui point and Dazhui point can inhibit the activity drop of brain mitochondria complex to maintain mitochondrial function in order to protect dopaminergic neurons.
