禽流感灭活疫苗H5N2 D7株与H5N1 Re-6株对鸭的免疫效果研究

摘要
为评价禽流感灭活疫苗(H5N2亚型,D7株)和重组禽流感病毒灭活疫苗(H5N1亚型,Re-6株)对鸭的免疫效果,根据兽医流行病学通过比较率判断疫苗应用效果的样本量计算方法,得出所需的最小样本量;以田间试验的方式,常规养殖白鸭300只、番鸭240只并进行随机分组,按程序免疫并在10日龄首免后2周开始,每周采集血清测定抗体效价直至出栏(白鸭45日龄,番鸭66日龄)。结果显示,10日龄白鸭和番鸭的母源抗体分别为3.02 log2和0 log2,需进一步开展研究以确定最佳的首免时间。首免后2周,无论是白鸭还是番鸭,H5N2 D7株疫苗组抗体HI效价和抗体合格率均高于H5N1Re-6株疫苗组;首免后3~5周,两种疫苗对白鸭和番鸭的HI效价均>4 log2,抗体合格率均≥70%。二免后1周至出栏,两种疫苗对白鸭和番鸭均产生较好的免疫效果,HI效价均>7 log2,抗体合格率均>80%。是否采用对应亚型的血凝抑制抗原对抗体的检测结果有较大的影响。研究表明,免疫两种疫苗后均能产生较好的免疫抗体,但相对而言,H5N2 D7株疫苗较H5N1 Re-6株疫苗更适合鸭的免疫,在鸭等水禽中推广使用该疫苗将更有效地防控水禽的高致病性禽流感。
To evaluate the immune efficacy of the inactivated avian influenza virus(AIV) H5N2 subtype vaccine(D7 stain) and H5N1 subtype vaccine(Re-6 stain) in ducks,300 white ducks and 240 muscovyducks were divided into 4 groups respectively,immunized with H5N2 D7 or H5N1 Re-6 inactivated vaccine with given immunization program and raised in normal way.Serum samples were collected from each group each week since two weeks after immunization till the time for marketing and then the hemagglutinin inhibition(HI) antibody was detected.The results showed that the time for first immunization for ducks may need further study as the maternal antibody titer of white ducks and muscovyducks were 3.5 log2 and 1.4 log2,respectively.Two weeks after the first immunization,the HI titers of H5N2 D7 vaccine groups were higher than that of H5N1 Re-6vaccine group both for white ducks and muscovyducks;The HI titers of two kinds of vaccines were all higher than 41og2 as the qualified rates were higher than 70%both for white ducks and muscovyducks 3-5 weeks after the first immunization.Since 1 week after the second immunization till time for marketing,the two vaccines produced good immune effect with antibodies HI titer all>71og2 and qualified rates> 80%in the two kinds of ducks.Besides,whether to adopt the corresponding antigen subtypes of HI will have some impact on the result of the antibody detection.This study concludes that although the two kind of vaccines could produce relatively good antibody titers,the H5N2 vaccine(D7 stain) is more suitable for ducks and may play more important role for prevention and controlling of HPA1 in waterfowl in China.
