香菇富锌培养及纤维素酶的研究

摘要
香菇富含丰富的蛋白质,维生素以及营养物质,本文以香菇为载体进行富锌培养,选用香菇早丰8号菌种,在PDA培养基中添加不同浓度梯度的外源锌对香菇进行培养,分析锌在香菇富集及生长过程中的影响,进行菌落大小观察、锌含量检测以及胞外酶酶活力的测定。试验结果表明:一定浓度的锌离子能促进香菇菌丝生长。硫酸锌添加量为700mg/kg的固体PDA培养中,香菇菌落直径最大。液体PDA培养中当硫酸锌添加量为900mg/kg液体培养时,香菇生物量达到最高,为2.68g/100mL。液体PDA培养基锌浓度在800mg/L时菌丝体锌含量最高,达到4.1451mg/g。菌包栽培时,在900mg/L的锌浓度时,菌丝体锌含量最高,为0.5611mg/g。固体栽培培养菌丝体中纤维素酶酶活性的测量中,纤维素酶活性综合比较好的是700mg/L锌浓度培养下的菌丝体,纤维素酶活呈现先上升后下降的趋势。
The Lentinus edodes is rich in protein,vitamins and nutrients.In this paper,as a carrier for Lentinus edodes cultivation in zinc,select and use Sawdust Lentinus edodes pack and Caofeng No.8 strain.Lentinus edodes were cultured in PDA Medium added different concentrations of exogenous zinc,Analysis of the effect of zinc in Lentinus edodes enrichment and growth,colony size observation,detection and determination of zinc content of ectoenzymes.The test results showed that:the zinc ion concentration can promote the growth of Lentinus edodes mycelium.The amount of training for 700mg/L zinc sulfate added in solid PDA,Lentinus edode's colony diameter is bigger than others.When the adding amount of 900mg/kg zinc sulfate in liquid PDA culture,Lentinus edodes biomass reached the highest,which is 2.68mg/100 mL.The content of zinc in the Liquid PDA cultured mycelia of 800mg/L was the highest in Liquid culture,which reached 4.1451mg/g.Dry cultivation of 900mg/L in zinc concentration in the high content of zinc,which is 0.5611mg/g.Dry cultivation mycelium Lentinus edodes enzymes activity measurements,cellulase activity comprehensive comparative better is 700mg/L zinc concentration in culture of mycelium,cellulase activity showed a trend Line increased and then decreased.
