牛源金黄色葡萄球菌随机扩增多态性分型

摘要
利用引物AP7建立随机引物多态性扩增体系对176株引起我国不同地区奶牛乳房炎的金黄色葡萄球菌分离株进行基因分型研究。结果表明,176株金黄色葡萄球菌均得到清晰的RAPD指纹图谱,扩增产物为2~9条带,产物大小为240~4500bp。菌株共分为12个基因型,其中Ⅰ型35株(20%);Ⅱ型和Ⅷ型各7株(各4%);Ⅲ型和Ⅺ型各6株(各3.4%);Ⅳ型17株(9.8%);Ⅴ型4株(2.2%);Ⅵ型43株(25%);Ⅶ型各12株(6.9%);Ⅸ型和Ⅻ型各9株(5.2%);Ⅹ型3株(1.7%)。Ⅰ型和Ⅵ型菌株占总菌株数45%以上,为主要流行的优势菌群。各地区基因型菌株分布比例有明显差异,可能与奶牛养殖业水平和环境差异有关。
Randomly amplified polymorphic DNA with three oligonucleotide primers were used for research on genetic polymorphism of 176 Staphylococcus aureus(S.aureus) strains isolated from dairy cattle in different regions of china.The result shows that all of the 176 S.aureus strains produced fingerprint distinctly and amplified out 2~9 electrophoresis strips which reveal DNA markers with sizes ranging from 240~4500bp.The S.aureus strains were separated into 12 genotype by fingerprint with oligonucleotide primers AP7 and 35 strains were type Ⅰ(20%),7 strains were each typeⅡ and Ⅷ(4%),6 strains were each type Ⅲ and Ⅺ(3.4%),17 strains were type Ⅳ(9.8%),5 strains were type Ⅴ(2.2%),43 strains were type Ⅵ(25%),12 strains were type Ⅶ(6.9%),9 strains were each type Ⅸ and Ⅻ(5,.2%),3 strains were type Ⅹ(1.7%).Difference of dairy farming level,geographical environment and using of antibiotic may be the reason of distinct discrepancy on proportion of different genotype strains in those areas.
