石斑鱼eIF2α基因PKR的克隆表达和功能初步分析

摘要
鱼类eIF2α激酶PKR与鱼类的免疫系统密切相关。本研究利用RACE技术,从石斑鱼的脾组织中克隆获得了PKR基因全长cDNA。通过构建原核表达载体PKR-p ET-32a,得到了PKR的融合蛋白,并将其用于制备了PKR的鼠抗血清。对PKR进行了组织分布研究石斑鱼的各组织中均有PKR的表达,石斑鱼的肠道和胃中PKR基因的表达量最高。对PKR进行了亚细胞定位研究,确定了PKR主要分布在细胞质中。用病毒诱导了石斑鱼细胞研究了PKR在病毒刺激下的表达量变化病毒后石斑鱼脑细胞中的PKR表达量相对于对照组均有不同程度的上升,其中VNN刺激下的PKR的表达量均缓缓升高大概在12小时处达到最高,在SGIV的刺激下PKR的表达量也是缓缓升高大概在48小时出表达量最高。研究表明,石斑鱼eIF2α激酶PKR在宿主抵御病毒感染的免疫反应中发挥了重要作用。
Fish eIF2 alpha kinase PKR is closely related to the immune system of fish. By constructing prokaryotic expression vector PKR-p ET-32 a, obtained the PKR fusion protein, and use it to mouse antiserum of PKR was prepared.Tissue distribution was studied for the PKR grouper of the PKR expression in each group, and PKR gene expression in the intestines and stomach grouper quantity highest. Has carried on the subcellular localization research to PKR,determines the PKR are mainly distributed in the cytoplasm. With the grouper cells induced PKR was studied under the virus to stimulate the expression of volume change after virus grouper brain cells relative to the amount of PKR expression in the control group have different degree rise,including under the stimulus of VNN, the expression of the PKRis slowly rising in about 12 hours to reach the highest, under the stimulus of SGIV the expression of the PKR is also slowly rising in about 48 hours with highest expression quantity.It is shown by this study that eIF2 alpha kinase PKR in host immune response against viral infections has played an important role in the grouper.
