珍珠提取物Ⅱ对人皮肤成纤维细胞增殖及胶原蛋白合成的影响

摘要
目的研究珍珠提取物Ⅱ对人皮肤成纤维细胞增殖及胶原蛋白Ⅰ型前蛋白合成的影响。方法通过培养获得人类正常真皮成纤维母细胞的融合单细胞,pNPP测试法评估细胞生存能力;酶联免疫吸附测定法量化胶原蛋白Ⅰ型前蛋白;Bradford法量化蛋白。结果活性成分"珍珠提取液Ⅱ"不会显著地降低人类正常真皮成纤维母细胞的生存能力,在添加量为0.1%、0.5%、1%时,均能提高成纤维母细胞合成胶原蛋白Ⅰ型前蛋白的能力。结论"珍珠提取液Ⅱ"具有显著提高人类正常真皮成纤维母细胞模型中合成胶原蛋白Ⅰ型前蛋白的功效。
Objective To evaluate the effect of the active compound Pearl Extract Ⅱ on the type Ⅰ procollagen production in a human normal dermic fibroblasts model.Methods By culturing cells of a 68 years old subject,the cellular viability was assessed by quantifying the intracellular phosphatases activity;the type Ⅰ procollagen was quantified in incubation media using a specific ELISA assay;proteins were quantified in the cell lysates by a Bradford method.Results Pearls Extract Ⅱ didn't reduced the viability of normal human dermic fibroblasts,and it was able to significantly increase the fibroblast type Ⅰ procollagen production when the additive is 0.1%,0.5%,and 1%.Conclusion the active compound Pearl Extract Ⅱ was able to significantly increase the fibroblast type Ⅰ procollagen production.
