摘要
目的:研究枳菊解郁汤对慢性不可预见性温和应激(CUMS)小鼠抑郁样行为的作用及其机制。方法:采用CUMS方式28天建立抑郁小鼠模型,灌胃枳菊解郁汤,通过开场、悬尾实验检测小鼠抑郁样行为变化;并且检测小鼠前脑皮层海马血管内皮生长因子(VEGF)的表达。结果:与对照组比较,模型组小鼠的水平活动度和自发活动明显减弱,总僵住不动时间明显延长;小鼠海马及前脑皮层VEGF表达水平明显降低;枳菊解郁汤连续灌胃28后,各剂量组和氟西汀组小鼠比模型组各检测指标均得到明显逆转。枳菊解郁汤中剂量组效果显著。结论:中药枳菊解郁汤可通过调控VEGF水平改善CUMS小鼠的抑郁样行为。
        Objective : To investigate the improvement and its mechanism of Zhijujieyu-tang on depression-like behaviors of chronic unpredictable mild stress( CUMS) mice,. Methods : The depression-like mice model was established by CUMS for 28 d. Zhijujieyu-tang and fluoxetine were intragastrically given to the mice. The depression-like behaviors were tested using the open field and tail suspension test. The changes of the Vascular endothelial growth factor( VEGF)in the hippocampus( HP) and prefrontal cortex( PFC) were evaluated by immunohistochemistry. Results : Compared with the control group, the exploration and spontaneous activity were significantly reduced, the immobility time were significantly increased, and the levels of VEGF contents in HP and PFC were apparently decreased in model group.After the mice were treated with fluoxetine and Zhijujieyu-tang, all the above indicators were significantly reversed.Zhijujieyu-tang medium dose group mice showed that it significantly effective. Conclusions : Zhijujieyu-tang improves behaviors of depressed model mice by modulating VEGF in HP and PFC.
