摘要
目的观察HOX转录反义RNA(HOTAIR)在非小细胞肺癌(NSCLC)组织中的表达变化,并探讨其临床意义。方法收集125例NSCLC患者的肿瘤组织及其瘤旁正常肺组织,采用实时RT-PCR法检测HOTAIR的表达。检索高通量基因表达数据库(GEO)、Array Express数据库中NSCLC组织与HOTAIR表达相关的芯片数据。结果125例患者中,HOTAIR在NSCLC组织中的表达高于癌旁正常肺组织(P<0.05)。HOTAIR表达水平与NSCLC患者年龄、肿瘤直径、淋巴结转移、TNM分期有关(P均<0.05)。从数据库检索出的基因芯片显示HOTAIR表达水平与肿瘤病理类型不相关(P>0.05)。结论 HOTAIR在NSCLC组织中呈现高表达,可能参与NSCLC的发生发展。
        Objective To observe the expression changes of HOX transcript antisense RNA( HOTAIR) in the nonsmall-cell lung cancer( NSCLC) tissues and to investigate its clinical significance. Methods NSCLC tissues and adjacent tissues from 125 patients were collected and the expression of HOTAIR was detected by RT-PCR. Microarray data on the correlations between HOTAIR expression and NSCLC tissues were retrieved from Gene Expression Omnibus( GEO) and Array Express database. Results The expression of HOTAIR in the NSCLC was significantly higher than that in the adjacent tissues( P < 0. 05). The expression of HOTAIR was closely related to age,tumor diameter,lymph node metastasis and TNM stage of NSCLC patients( all P < 0. 05). The microarray data from GEO Profiles demonstrated that the HOTAIR expression level was not related with pathological pattern( P > 0. 05). Conclusion HOTAIR is highly expressed in the NSCLC tissues,and its expression may take part in the occurrence and development of NSCLC.
