血管通透因子、DCC基因及MMP-9蛋白在胃癌患者中的表达水平研究

摘要
目的:研究血管通透因子(vascular permeability factor,VPF)、结直肠癌缺失(deleted in colorectal cancer,DCC)基因及基质金属蛋白酶9(matrix metalloprotein-9,MMP-9)蛋白在胃癌患者中的表达水平及意义。方法:取50例胃癌患者手术切除的癌组织以及与癌组织距离5 cm以上的癌旁正常胃黏膜组织制作标本,使用酶联免疫吸附法检测VPF水平,荧光定量RT-PCR检测DCC基因水平,Western blot检测MMP-9蛋白表达水平。结果:癌组织中VPF、MMP-9阳性表达率显著高于癌旁组织,DCC基因阳性表达率显著低于癌旁组织(P<0.05);胃癌患者VPF、DCC及MMP-9阳性表达率与性别、年龄无关,与病理分期、淋巴结转移情况、肿瘤体积、浸润程度以及分化程度有关;Ⅲ、Ⅳ期、有淋巴结转移、肿瘤体积较大、侵及浆膜以及分化程度较高的患者VPF、MMP-9阳性表达率较高,DCC基因阳性表达率较低(P<0.05)。结论:血清VPF、DCC基因及MMP-9水平在胃癌症状中呈现异常表达,且具有一定相关性,为胃癌的临床诊断提供一定的价值帮助。
Objective:To investigate the expression and significance of vascular permeability factor(VPF),deleted in colorectal cancer(DCC) gene and matrix metalloproteinase-9(MMP-9) in gastric cancer.Methods:The specimens of 50 cases of gastric cancer resected surgically and normal gastric mucosa adjacent to the cancer more than 5 cm away from the cancer tissue were prepared,using enzyme-linked immunosorbent method to detect VPF,using fluorescence quantitative RT-PCR to detect DCC gene level,using Western blot to detect MMP-9 protein expression levels.Results:Positive expression rates of VPF and MMP-9 in cancer tissues were significantly higher than those in paracancerous tissues,and positive expression rates of DCC genes were significantly lower than those in paracancerous tissues(P<0.05).The positive expression rates of VPF,DCC and MMP-9 in gastric cancer patients were not related to sex and age,but related to pathological stage,lymph node metastasis,tumor volume,infiltration degree and differentiation degree.The positive rates of VPF and MMP-9 were higher in patients with stage III and IV,lymph node metastasis,larger tumor volume,serosa invasion and higher differentiation,while the positive rates of DCC gene were lower(P<0.05).Conclusion:Serum levels of VPF,DCC gene and MMP-9 are abnormally expressed in gastric cancer symptoms,and have a certain correlation,providing certain value for clinical diagnosis of gastric cancer.
