摘要
目的观察丝胶对2型糖尿病(T2DM)大鼠胰岛β细胞胰岛素蛋白表达的影响。方法 36只雄性SD大鼠随机分为正常对照组、糖尿病模型组和丝胶治疗组,每组12只大鼠。链脲佐菌素腹腔注射建立T2DM大鼠模型并给予丝胶(2.4 g/kg)灌胃。SP免疫组织化学染色法观察大鼠胰岛细胞胰岛素蛋白的表达情况。结果胰岛素蛋白免疫阳性产物位于胰岛β细胞胞质,呈棕黄色颗粒状。糖尿病模型组大鼠胰岛β细胞胰岛素蛋白的表达水平为(0.392±0.017),明显低于正常对照组大鼠(0.729±0.017)(P<0.01);丝胶治疗组大鼠胰岛β细胞胰岛素蛋白的表达水平为(0.619±0.039),明显高于糖尿病模型组大鼠(P<0.01)。结论丝胶可通过上调胰岛素蛋白的表达保护糖尿病时胰岛β细胞损伤。
        Objective To observe the effects of sericine on insulin protein expression in islet cells of type 2 diabetes mellitus rats. Methods 36 male SD rats were randomly divided into 3 groups with 12 rats in each group: normal control,diabetes mellitus model and sericine treatment groups. Type 2 diabetes mellitus rat model was made by continuous intraperitoneal injection of streptozotocin,and then the rats were lavaged with sericine( 2. 4 g / kg). SP immunohistochemical stain was used to observe insulin protein expression in islet cells. Results Insulin protein positive immunoreactive products were brown and yellow granules and located in cytoplasm of islet beta cells. Compared with normal control rats( 0. 729±0. 017),the insulin protein expression in beta cells of rats in diabetes mellitus model group( 0. 392±0. 017) obviously was decreased( P<0. 01). The insulin protein expression in beta cells of rats in sericine treatment group( 0. 619±0. 039) was obviously higher than that of diabetes mellitus model group( P<0. 01). Conclusions Sericine can protect islet beta cells by up regulating insulin protein expression during diabetes mellitus.
