摘要
目的探讨涤痰汤干预阿尔茨海默病(Alzheimer’s disease, AD)模型大鼠的效果及机制。方法通过对Sprague Dawley大鼠双侧海马注射冈田酸(okadaic acid, OA)复制AD大鼠模型,用涤痰汤水煎液[1 g/100g/d]灌胃,3周后利用Morris水迷宫和Western blotting分别检测AD模型大鼠空间学习记忆能力和tau、PP2Ac蛋白磷酸化水平的变化。结果涤痰汤干预后能显著逆转OA诱导的大鼠AD样学习记忆能力减退,并能明显减轻OA诱导的tau蛋白Ser214、Ser262位点以及PP2Ac Tyr307位点的过度磷酸化。结论涤痰汤可逆转OA引起的AD样模型大鼠的学习记忆能力衰退,其机制可能与激活PP2A从而减轻tau蛋白的过度磷酸化有关。
