海参肽对D-半乳糖衰老模型小鼠的抗衰老作用研究

摘要
目的研究海参肽对D-半乳糖致衰老小鼠抗氧化和免疫功能的影响。方法将60只BALB/c小鼠,分为正常组(给予生理盐水)、模型组(给予生理盐水)、海参肽低、中、高三个剂量组(125、250、500 mg/kg)和VE组(100mg/kg),采用颈背部皮下注射1 000 mg/kg剂量的D-半乳糖来建立小鼠衰老模型,记录小鼠的体重变化,计算胸腺和脾脏指数,测定血清谷胱甘肽过氧化物酶(GSH?Px)活力、脂褐素(Lipo)含量、免疫球蛋白G(IgG)、白细胞介素-2(IL?2)含量、心肝组织中超氧化物歧化酶(SOD)活力和Lipo含量。结果与模型组相比,海参肽中、高剂量组活力、心肝组织的SOD活力,降低血清Lipo含量,差异均有统计学意义(P<0.05);海参肽高剂量组(500 mg/kg)能升高血清免疫因子IgG、IL?2含量,降低心脏组织的Lipo含量,差异有统计学意义(P<0.05)。结论海参肽对D-半乳糖诱导的衰老模型小鼠具有提高免疫和增强抗氧化能力的作用,能够延缓衰老。
        Objective To study the effect of sea cucumber peptide on immune and anti?oxidation function of aging mice induced by D?galactose. Methods 60 BALB/c mice were randomly divided into normal group,model,sea cucumber peptide low?dose,medium?dose and high?dose groups(125,250,500 mg/kg)and VE group(100 mg/kg). The aging model of mice was established by subcutaneous injection of high dose(1 000 mg/kg)D?galactose at the neck. The weight changes of mice were recorded. After 8 weeks,the spleen index,the thymus gland index,the activity of glutathione peroxidase(GSH?Px)and the level of immunoglobulin G(IgG),interleukin?2(IL?2)in serum,the activity of superoxide dismutese(SOD)and the content of lipofuscin(Lipo)in heart and liver tissues were detected. Results Compared with the model group,the index of thymus and spleen,the activity of GSH increased in medium and high dose groups(250,500 mg/kg)and the content of Lipo was significantly decreased in sea cucumber peptide medium and high dose groups(250,500 mg/kg)(P<0.05). Compared with the model group,the content of IgG,IL?2 in serum was significantly increased and the content of Lipo in heart tissue was significantly decreased in high dose group(500 mg/kg)(P<0.05). Conclusion Sea cucumber peptide could improve immunity and anti?oxidation ability of D?galactose induced aging model mice,suggesting that it might be effective in anti?aging process.
