香茅草黄酮提取物对L-NNA诱导的高血压大鼠血压及炎性因子的影响

摘要
目的探讨香茅草黄酮提取物对左旋硝基精氨酸(L-NNA)诱导的高血压大鼠血压的影响及其作用机制。方法采用N,-硝基-L-精氨酸(L-NNA)15 mg/(kg·d)腹腔注射给药4周复制高血压动物模型,选择平均动脉压高于140 mmHg的大鼠50只进行实验,将其随机分为5组,即模型组、香茅草黄酮提取物低剂量、中剂量、高剂量组、卡托普利阳性药物组,另外选取正常10只大鼠作为空白组。模型组和空白对照组给予同等体积生理盐水;阳性药物对照组大鼠给予1%卡托普利2 mL/d;香茅草黄酮提取物低(0.85%)、中(1.65%)、高(3.35%)浓度剂量组给予香茅草黄酮提取物2 mL/d,各组连续灌胃给药4周,每周测定大鼠尾动脉血压。4周后,采用开腹腔后,用真空采血管于腹主动脉取血,用ELISA法检测血清中肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)的水平。结果给药后,香茅草黄酮提取物低、中、高剂量组及阳性药物对照组与模型组比较,呈现明显的降压作用(P <0.05)。与空白组相比,模型组大鼠血清中TNF-α,IL-6含量显著升高(P <0.05)。4周后,香茅草黄酮提取物中、高剂量组以及卡托普利阳性药物组大鼠血清中TNF-α、IL-6含量低于模型组,差异有统计学意义(P <0.05)。结论香茅草黄酮提取物对L-NNA诱导的高血压有一定的降压作用,并能有效降低高血压的炎症水平,其降压机制可能与炎性因子有关,具体的抗炎机制还有待于进一步探究。
        Objective To investigate the effect of flavonoids extract from cymbopogon on blood pressure induced by L-NNA in hypertensive rats, and to study the mechanism of its effect on blood pressure. Methods Rats with N'-nitro-L-arginine(L-NNA) 15 mg/(kg·d) were administered intraperitoneally for four weeks to replicate the animal model of hypertension. 50 rats with an average arterial pressure higher than 140 mmHg were selected for the experiment and randomly divided into five groups, namely model group, cymbopogon low dose, medium dose and high dose group,captopril positive drug group, and another ten normal rats were selected as the blank group. The model group and the blank control group were given the same volume physiological saline. The rats in the positive control group were given1% captopril 2 mL/d; low(0.85%), medium(1.65%) and high(3.35%) concentration dose group were given 2 mL/d of cymbopogon extract, and each group was given a continuous gavage for four weeks, and the arterial blood pressure was measured twice a week and the serum levels of TNF-α and IL-6 were measured by ELISA. Results Compared with the model group, cymbopogon low dose, medium dose and high dose group, captopril positive drug group showed obvious hypotensive effect(P < 0.05). Compared with the blank group, the serum TNF-α and IL-6 in the model group were significantly increased(P < 0.05). After four weeks, the serum TNF-α and IL-6 in the medium and high dose group and captopril group were lower than those in the model group(P <0.05). Conclusion Cymbopogon extracts of L-NNA induced hypertension has certain function, and can reduce the inflammation level of hypertension effectively. Its function mechanism may be related to inflammatory factors, and the specific anti-inflammatory mechanism remains to be further explored.
