摘要
目的:探讨支气管哮喘患儿外周血T细胞亚群、血清趋化因子及酶类多肽水平变化及意义。方法:60例支气管哮喘患儿为研究组,另选取同期体检健康儿童50例为对照组,采集2组受试者清晨空腹静脉血3 mL,使用流式细胞仪检测外周血CD+4T细胞亚群中辅助性T淋巴细胞(Th1、Th2、Th17)及调节性T细胞(Treg),计算Th1/Th2、Th17/Treg比值;另采集清晨空腹静脉血3 mL,采用酶联免疫吸附法(ELISA)检测血清白细胞介素-2(IL-2)、IL-4、IL-6、IL-17、干扰素-γ(IFN-γ)、嗜酸性粒细胞趋化因子(Eotaxin)、巨噬细胞衍生趋化因子(MDC)、单核细胞趋化蛋白-1(MCP-1)、单核细胞趋化蛋白-4(MCP-4)、基质金属蛋白酶-1(MMP-1)、基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶-9(MMP-9)、组织蛋白酶K(Cat K)、组织金属蛋白酶抑制因子-1(TIMP-1)水平。结果:研究组外周血Th1、Treg、Th1/Th2低于对照组,Th2、Th17、Th17/Treg高于对照组(P<0.05);研究组血清IL-2、IFN-γ水平低于对照组,IL-4、IL-6、IL-17水平高于对照组(P<0.05);研究组血清Eotaxin、MDC、MCP-1、MCP-4水平高于对照组(P<0.05),研究组血清MMP-1、MMP-2、MMP-9、Cat K水平高于对照组、TIMP-1水平低于对照组(P<0.05)。结论:支气管哮喘患儿发病与外周血T淋巴细胞亚群、酶类多肽、趋化因子异常表达有关。
        Objective:To explore the changes and significance of T cell subsets in peripheral blood,serum chemokines and enzyme peptides in children with bronchial asthma.Methods:60 children with bronchial asthma were selected as study group and 50 healthy children as control group.Fasting venous blood was collected from two groups for 3 mL in the morning.Auxiliary T lymphocytes(Th1、Th2、Th17) and regulatory T cells(Treg) in Peripheral Blood CD4+T Cell subsets were detected by flow Cytometry to calculate the ratio of Th1/Th2 and Th17/Treg.After 3 mL of fasting venous blood was collected in the morning,serum Interleukin(IL)-2(IL-2),IL-4,IL-6,IL-17,IFN-γ,Eotaxin,MDC,MCP-1,MCP-4,MMP-1,MMP-2,MMP-9,Cat K and TIMP-1 were detected by Enzyme-linked Immunosorbent Assay(ELISA).Results:Th1,Treg,and Th1/Th2 in the study group were significantly lower than that in the control group,while Th2,Th17,and Th17/Treg were significantly higher than that in the control group(P<0.05).IL-2 and IFN-γ in the study group were significantly lower than that in the control group,while IL-4,IL-6,and IL-17 were significantly higher than that in the control group(P<0.05).Eotaxin,MDC,MCP-1,and MCP-4 in the study group were significantly higher than that in the control group(P<0.05).MMP-1,MMP-2,MMP-9,and Cat K in the study group were significantly higher than that in the control group,while TIMP-1 was significantly lower than that in the control group(P<0.05).Conclusion:The pathogenesis of bronchial asthma is related to the abnormal expression of T lymphocyte subsets,enzyme peptides and chemokines in peripheral blood.
