摘要
目的探讨川陈皮素对高血压大鼠的血压调节作用及分子机制。方法使用L-NNA制造内皮损伤型高血压大鼠模型,给予不同浓度的川陈皮素,分别在造模前、造模后、给药14天及给药28天时测量大鼠血压、心率和体重的改变。处死给药28天大鼠,取动脉血,使用ELISA法测量NO、EDHF、ANG-2和PGI2含量。结果川陈皮素给药14天及28天后,高血压大鼠血压明显下调,且ELISA结果显示高血压大鼠动脉血中NO和PGI2的含量增加,但EDHF和ANG-2含量无影响。结论川陈皮素可影响血管内皮细胞功能,提高NO和PGI2的含量,使高血压大鼠血压下降。
Objective To investigate role of nobiletin on the blood pressure regulation in hypertension rats and its mechanism. Methods The hypertensive rat model with endothelial injury was prepared by L-NNA and then was injected by the different contents of nobiletin. Changes of artery pressure, heart rate, and weight before and after model establishment at 14 days and 28 days after model establishment were detected. All of rats that had been injected for 28 days were killed and their artery plasma were collected. The contents of NO, EDHF, ANG-2 and PGI2 were measured. Results The blood pressure decreased obviously after treated with nobiletin for 14 days and 28 days. And ELISA results showed that the total NO and PGI2 contents in artery blood increased, but the PGI2 contents had no change. Conclusion Nobiletin may have an important effect on the function of the vascular endothelium and increase the PGI2 and NO contents, causing to lower artery pressure.
