摘要
目的建立快速、同时测定人血中环孢素A及伏立康唑血药浓度的方法。方法采用双波长高效液相色谱(HPLC)法,色谱柱为Diamonsil RP-C_(18)柱(150 mm×4.6 mm,5μm),流动相为乙腈-0.01 mol/L醋酸铵洗脱,检测波长环孢素A为215 nm,伏立康唑为256 nm,流速为1 mL/min。结果伏立康唑及环孢素A保留时间分别为5.32 min和6.48 min,血药浓度线性范围分别为30~1 500 ng/mL和200~10 000 ng/mL,准确度均在95.0%以上;日内、日间精密度的RSD均小于15%(n=5)。结论该方法灵敏、稳定,结果准确、可靠,可用于同时测定环孢素A及伏立康唑的血药浓度。
Objective To establish a method for rapid and simultaneous determination of blood concentration of cyclosporine A and voriconazole in human plasma.Methods Dual-wavelength HPLC was adopted,the chromatographic column was Diamonsil RP-C_(18) column(150 mm × 4.6 mm,5 μm),the mobile phase was acetonitrile-0.01 mol/L ammonium acetate elution,the detection wavelength was 215 nm for cyclosporine A,256 nm for voriconazole,and the flow rate was 1 mL/min.Results The retention time of voriconazole and cyclosporine A was 5.32 min and 6.48 min,respectively,their linear ranges of plasma concentration were 30-1 500 ng/mL and200-10 000 ng/mL,their accuracy was over 95.0%,and RSDs of intra-day and inter-day precision were less than 15%(n=5).Conclusion The method is sensitive,stable,accurate and reliable,which can be used for the simultaneous determination of blood concentration of cyclosporine A and voriconazole.
