海州常山花萼转色期生理变化研究

摘要
【目的】探究海州常山花萼色素的组成及动态变化规律。【方法】以分别从盐城亭湖、镇江句容、连云港云台山、徐州贾汪、泰安泰山和驻马店东官庄引种至南京的6个种源海州常山为试验材料,用显色反应和紫外-可见光谱检测海州常山花萼中色素的组成,运用HPLC法鉴定花青素组分,于盛花初期、盛花中期、花果共存期、结果期、果萼宿存期和落叶期采样测定花萼变色过程中叶绿素、花青素、可溶性糖含量和相关酶活性的变化,并分析花青素与其他指标的相关性。【结果】海州常山花萼中含有叶绿素、花青素和类黄酮化合物,不含类胡萝卜素。从不同发育期海州常山花萼中共检测出3种花青素成分,确定其中一种为矢车菊素-3-O-葡萄糖苷。在花萼变色过程中,叶绿素含量降低,花青素与可溶性糖含量升高,苯丙氨酸解氨酶(PAL)活性在转色后期波动较大,查尔酮异构酶(CHI)活性明显升高。相关性分析发现,花青素含量与叶绿素、可溶性糖含量和PAL、CHI活性明显相关。【结论】转色过程中CHI酶的变化是海州常山花萼转色的主要原因。
        【Objective】To explore the composition and dynamic changes of the calyx pigment in Clerodendrum trichotomum.【Method】Six Clerodendrum trichotomum samples(Tinghu,Yancheng;Jurong,Zhenjiang;Yuntaishan,Lianyungang;Jiawang,Xuzhou;Taishan,Tai'an;Dongguanzhuang,Zhumadian)from different provenances introduced into Nanjing were used as experimental materials to detect pigment composition in the calyx through identifying the anthocyanin components using color reaction,UV-VIS spectroscopy and HPLC method.At the six developmental stages(primary full-blooming stage,middle fullblooming stage,flower and fruit coexistence stage,fruit-bearing stage,fruit and calyx persistent stage,defoliating stage)sampling,mesuring the changes of chlorophyll,anthocyanin,soluble sugar and anthocyanidin synthase in the process of calyx discoloration,meanwhile analyzing the correlation between anthocyanins and other indicators.【Result】It showed that there were chlorophyll,anthocyanin and flavonoids,no carotenoids in the Clerodendrum trichotomum calyx.A total of 3 anthocyanin constituents were identified in which the Cyanidin-3-O-glucoside was confirmed.During the process of color change,the content of chlorophyll decreased,the content of anthocyanin and soluble sugar increased,PAL enzyme activity fluctuated greatly at the later color-turning stage and the activity of CHI enzyme increased obviously.Through correlation analysis found that anthocyanin content was significantly correlated with chlorophyll content,soluble sugar content,PAL and CHI enzyme activities.【Conclusion】Change in CHI enzyme during color conversion was the main cause of calyx color change in Clerodendrum trichotomum.
