摘要
以青钱柳(Cyclocarya paliurus)种胚萌发的无菌苗为外植体,研究植物生长调节剂对试管苗增殖及植株再生的影响,结果表明:较适宜青钱柳增殖的基本培养基为1/2MS培养基;最佳继代增殖培养基为1/2MS+3.0mg·L~(-1)6-苄氨基腺嘌呤(6-BA)+0.05mg·L~(-1)吲哚-3-乙酸(IAA),增殖系数可达7.5;最佳壮苗伸长培养基为1/2MS+0.5mg·L~(-1)IAA+5.0mg·L~(-1)赤霉素(GA_3);最佳生根培养基为1/2MS+0.5mg·L~(-1)吲哚-3-丁酸(IBA)+0.5mg·L~(-1) (萘乙酸) NAA,生根率可达32.0%。
        In order to establish the tissue culture system of Cyclocarya paliurus, the effects of plant growth regulators on proliferation and plant regeneration in vitro were studied using the sterile seedling sprouting from embryos as explants. The results show that the best suitable basic medium for sterile seedling of C. paliurus multiplication was 1/2 MS; the best medium for shoot multiplication was 1/2 MS+3.0 mg·L~(-1) 6-benzylaminopurine(6-BA)+0.05 mg·L~(-1) indole-3-acetic acid(IAA), and the multiplication coefficient can reach 7.5; the best medium for seedling elongation culture was 1/2 MS+0.5 mg·L~(-1) IAA+5.0 mg·L~(-1) gibberellin A_3(GA_3); the best rooting medium was 1/2 MS+0.5 mg·L~(-1) indole-3-butyric acid(IBA)+0.5 mg·L~(-1) 1-naphthylacetic acid(NAA), with a 32.0% rooting rate.
