野艾蒿提取物对金黄色葡萄球菌的抑菌机制研究

摘要
文章研究野艾蒿提取物对金黄色葡萄球菌(SA)的作用机制。通过野艾蒿提取物作用后SA细胞外碱性磷酸酶(AKP)含量、电导率大小以及DNA和RNA大分子物质含量的变化研究其对SA细胞壁及细胞膜通透性的影响;通过测定SA胞内蛋白含量的变化研究野艾蒿提取物对SA蛋白合成的影响;通过测定苹果酸脱氢酶(MDH)和琥珀酸脱氢酶(SDH)活性研究野艾蒿提取物对SA呼吸代谢的影响;通过扫描电镜观察野艾蒿提取物对SA细胞形态的影响。通过1%的葡聚糖凝胶电泳测定野艾蒿提取物与DNA的相互作用。结果表明,野艾蒿提取物作用后SA胞外AKP含量升高,电导率增加,核酸含量无明显变化,SA胞内蛋白含量降低,MDH和SDH活性降低,细胞形态无明显变化,野艾蒿提取物与SA的DNA发生紧密的结合。由此可知,野艾蒿提取物是通过影响SA细胞壁及细胞膜通透性、胞内蛋白含量、MDH和SDH活性并且与SA的DNA发生结合发挥抑菌作用。
        To study the action mechanism of Artemisia argyi extract on Staphylococcus aureus(SA). The effects of Artemisia argyi extract on SA cell wall and cell membrane permeability were studied by changing the content of extracellular alkaline phosphatase(AKP), conductivity and DNA and RNA macromolecule substances after the action of Artemisia argyi extract. The effects of Artemisia argyiextract on SA protein synthesis were studied by measuring the content of intracellular protein in SA. Malic acid dehydrogenase(MDH) and succinate dehydrogenase(SDH) were determined, these activity was used to study the effect of Artemisia argyi extract on respiratory metabolism of SA, and the effect of Artemisia argyi extract on SA cell morphology observed by scanning electron microscopy. The interaction between Artemisia argyi extract and DNA was detected by 1% dextran gel electrophoresis. The results showed that the content of extracellular AKP, conductivity in SA increased, the content of intracellular protein in SA decreased, the activity of MDH and SDH decreased nucleic acid, and the cell morphology did not change significantly after treated with Artemisia argyi extract. Artemisia argyi extract was closely bound to the DNA of SA. It can be concluded that Artemisia argyi extract exerts bacteriostasis by affecting SA cell wall and cell membrane permeability, intracellular protein content, MDH and SDH activity, and binding with SA DNA.
