摘要
目的研究全蝎白术蛴螬混合发酵品(SAHHFP)体内外抗呼吸道合胞病毒(RSV)的药理作用。方法将SAHHFP用不同极性的溶剂、不同的提取方式提取,分别得到不同的极性部位,用MTT法进行体外抗RSV病毒实验,通过比较抗病毒治疗指数TI来确定最佳提取方法;体内抗RSV病毒实验,用RSV病毒感染小鼠模型,通过计算胸腺指数、肺指数、酶联免疫试剂盒检测血清中肿瘤坏死因子(TNF-α)、γ-干扰素(IFN-γ)、一氧化氮(NO)三种细胞因子的含量等来观察体内抗病毒效果。结果体外抗病毒方面,SAHHFP 50%超声醇提液抗病毒效果最好;体内抗病毒方面,SAHHFP高中低三个剂量的肺指数抑制率以及三种细胞因子的含量均高于模型组。结论 SAHHFP 50%超声醇提液对RSV病毒在体外和体内都有较好的抑制作用,且和模型组相比,具有显著性差异。
        Objective To study the pharmacological effects of mixed fermentation of the Scorpion and Atractylodes Macrocephala Rhizome and Holotrichia Larva(SAHHFP) against respiratory syncytial virus(RSV) in vitro and in vivo.Methods SAHHFP was extracted with different polar solvents and different extraction methods to obtain different polar sites. It was performed by MTT method, and the optimal extraction method was determined by comparing the antiviral therapeutic index TI of in vitro anti-RSV experiment.In vivo, mouse model was infected with RSV virus, and the antiviral effects were detected by calculating thymus index, lung index,and the contents of three cytokines such as serum tumor necrosis factor(TNF-α),γ-interferon(IFN-γ),nitrogen(NO). Results In vitro, The 50 % alcohol extraction of SAHHFP had better anti-RSV virus effect, and of which,50 % ultrasonic alcohol extract had the best antiviral effect; in vivo, the lung index inhibitions and the contents of the three cytokines of the high, middle and low doses were higher than the model group.Conclusion The 50% ultrasonic alcohol extract of SAHHFP has a good inhibitory effects on RSV virus in vitro and in vivo, and has significant difference comparing with the model group.
