摘要
目的:研究小儿解感颗粒体内外对呼吸道合胞病毒(RSV)感染的影响及意义。方法:体外实验用人喉癌细胞(Hep-2)培养法,以治疗指数(TI)为研究指标,以利巴韦林为阳性对照药,研究不同剂量小儿解感颗粒对RSV的抑制作用;体内实验选用BALB/c小鼠,运用肺指数检测法、HE染色法、ELISA检测法研究该药体内抗RSV作用。结果:体外实验,小儿解感颗粒TC50=1.65 mg/mL,TC0=1.25 mg/mL,TI=5.06;体内实验,与模型组相比,小儿解感颗粒高、中剂量组肺指数显著下降(P<0.05);HE染色观察得出小儿解感颗粒高剂量组与正常组形态最为接近;ELISA检测得出,用药组白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)含量随剂量的增大而减少,且均比利巴韦林组效果显著,小儿解感颗粒高剂量组TNF-α含量明显降低(P<0.05)。结论:小儿解感颗粒体外抑制RSV作用明显,体内对RSV引起的疾病有明显治疗作用。
        Objective:To study the effect and significance of anti-respiratory syncytial virus(RSV) of Xiaoer Jiegan Granules in vitro and in vivo. Methods:In vitro experiments,using human laryngeal cancer cells(hep-2) culture method,with the treatment index(TI) as the research index,elibavirin as the positive control drug,the inhibitory effect of Xiaoer Jiegan Granules with different dosage on RSV was studied. BALB/c mice were selected for the in vivo experiment,and the anti-RSV effect of this drug in vivo was studied by lung index detection method,HE staining method and ELISA detection method. Results:In vitro,Xiaoer Jiegan Granules TC0=1.25 mg/mL,TC50=1.65 mg/mL,TI=5.06.In vivo,compared with the model group,the lung index of Xiaoer Jiegan Granules high and medium dose groups decreased significantly(P <0.05). HE staining method showed that the morphology of Xiaoer Jiegan Granules high-dose group was the closest to that of the normal group. ELISA detection method showed that the content of IL-6 and TNF-αin the treatment group decreased with the increase of dose,and the effect was significantly higher than that in the bavirin group,while the content of TNF-α in Xiaoer Jiegan Granules high-dose group decreased significantly(P<0.05). Conclusion:Xiaoer Jiegan Granules has obvious inhibitory effect on RSV in vitro,and it has obvious therapeutic effect on RSV-induced diseases in vivo.
