痛风二号方对湿热型高尿酸血症大鼠血清肿瘤坏死因子α和血管活性肠肽水平的影响研究

摘要
目的:研究湿热型高尿酸血症(HUA)大鼠血尿酸、肿瘤坏死因子α(Tnf-α)和血管活性肠肽(VIP)的变化,以及痛风二号方对HUA大鼠血清尿酸、肿瘤坏死因子α、血管活性肠肽的影响。方法:将40只Wistar雄性大鼠,随机分为空白对照组、湿热高尿酸血症模型组(模型组)、别嘌醇组、痛风二号方组。空白对照组:正常喂饲,在其余三组进行模型建立和药物干预的时候予等量生理盐水行灌胃、腹腔注射等相应操作。湿热型高尿酸血症模型组:高脂高糖饮食、白酒灌胃、200g/L蜂蜜水自由饮用,持续14天。自第八天开始,酵母膏按比例混入高脂饲料喂饲联合腹腔注射氧嗪酸钾,制备大鼠模型;同时自第10天开始,将大鼠置入湿热环境中放置4天。痛风二号方组,别嘌醇组:造模同湿热型高尿酸血症模型组,自造模第八天起,灌胃及注射造模药物后6小时,分别给予大鼠痛风二号方(0.3g/kg)或别嘌醇(40mg/kg)灌胃干预,直到第14天。生化分析仪检测大鼠血清尿酸值;通过ELISA法检测大鼠血清VIP、Tnf-α。结果:模型组、两药物干预组(别嘌醇组、痛风二号方组)血尿酸值、血清Tnf-α、血清VIP值与空白对照组相比,差异均具有显著性统计学意义(P<0.01),痛风二号方组血尿酸值、血清Tnf-α、血清VIP值与模型组相比差异具有显著性统计学意义(P<0.01),别嘌醇组与模型组血尿酸值、血清Tnf-α差异具有显著性统计学意义(P<0.01或P<0.05),血清VIP差异无统计学意义(P>0.05),两药物干预组之间血尿酸值、血清Tnf-α、血清VIP值差异具有显著统计学意义(P<0.01或P<0.05)。结论:痛风二号方对湿热型高尿酸血症有明显的治疗作用,在降尿酸的同时还可降低Tnf-α及升高VIP,且疗效优于别嘌醇,对湿热型高尿酸血症有较好的防治作用。
        Objective:To study the changes of serum uric acid,tumor necrosis factor-α(Tnf-α) and vasoactive intestinal peptide(VIP) in(HUA) rats with hyperuricemia of damp-heat type,and the effect of Tongfeng No.2 Recipe on serum uric acid and Tnf-αin HUA rats,and effect of vasoactive intestinal peptides.Methods:Forty male Wistar rats were randomly divided into blank controlled group,model group with hyperuricemia of dampness and heat,allopurinol group and Tongfeng No.2 Recipe group.The blank controlled group:normal feeding,the other three groups of model and drug intervention with the same amount of normal saline,intraperitoneal injection and other corresponding operations.Hygrothermal hyperurination acidemia model group:high fat and high sugar diet,liquor by stomach,200 g/L honey water for 14 days.From the eighth day,the rats were fed with high fat diet and intraperitoneal injection of potassium oxazinate,and then placed in humid and hot environment for 4 days.Tongfeng No.2 Recipe group,allopurinol group:the model group of hyperuricemia of the same damp-heat type,from the eighth day of the model,6 hours after intragastric administration and injection of the drug to make the model.Rats were given Tongfeng No.2 Recipe(0.3 g/kg) or allopurinol(40 mg/1 kg) respectively,until the Fourteenth days.Serum uric acid was measured by biochemical analyzer and serum VIP,Tnf-αwas detected by ELISA method.Results:There were significant differences in serum uric acid,serum Tnf-α and serum VIP between the two drug intervention groups(the allopurinol group,the Tongfeng No.2 Recipe group) compared with the blank controlled group(P<0.01).The levels of serum uric acid,serum Tnf-α and serum VIP in Tongfeng No.2 Recipe group were significantly different from those in the model group(P<0.01),and the serum uric acid value in the allopurinol group and the model group was significantly higher than that in the model group(P<0.01).There was significant difference in serum Tnf-α(P<0.01 or P<0.05),but no significant difference in serum VIP(P>0.05) there were significant differences in serum uric acid,serum Tnf-αand serum VIP between the two drug intervention groups(P<0.01 or P<0.05).Conclusion:Tongfeng No.2 Recipe has obvious therapeutic effect on hygrothermal hyperuricemia.It can reduce Tnf-αand increase VIP,at the same time,and the curative effect is better than allopurinol.It has better preventive and therapeutic effect on hygrothermal hyperuricemia.
