摘要
目的观察吲哚-2,3-二酮对醋酸棉酚致雄性小鼠生精障碍模型生精功能的影响。方法雄性昆明种小鼠分为正常对照组、模型对照组和吲哚-2,3-二酮25、50、100 mg·kg~(-1)组(n=8)。醋酸棉酚20 mg·kg~(-1)灌胃30 d制备生精障碍模型。制模同时吲哚-2,3-二酮各剂量组灌胃给予相应剂量30 d后,观察小鼠精子、睾丸发育及生育的影响。结果与空白对照组比较,模型对照组精子密度和活动率均降低(P<0.01),精原细胞、初级精母细胞及精子细胞数目减少(P<0.01);吲哚-2,3-二酮25、50、100 mg·kg~(-1)组精子密度和活动率显著高于模型对照组(P<0.05或P<0.01),吲哚-2,3-二酮25 mg·kg~(-1)组初级精母细胞数目增多(P<0.05),50 mg·kg~(-1)、100 mg·kg~(-1)组精原细胞、初级精母细胞及精子细胞数目均增多(P<0.01),呈剂量依赖性。模型对照组孕鼠1只,胎鼠3只;吲哚-2,3-二酮25 mg·kg~(-1)、50 mg·kg~(-1)和100 mg·kg~(-1)组孕鼠分别为2、3、6只,胎鼠分别为(6.1±2.1)、(7.5±1.7)和(8.6±1.4)只。结论吲哚-2,3-二酮可改善醋酸棉酚致雄性小鼠生精障碍模型的生精功能。
        AIM To investigate the effects of indole-2, 3-dione on the spermatogenesis impairment model induced by gossypol acetate on male mice. METHODS The male Kunming mice were divided into normal control group, model control group and indole-2, 3-dione 25, 50, 100 mg·kg~(-1) groups according to random digits table( n = 8). Except the normal control group, other groups were intragastricly administrated with gossypol acetate 20 mg·kg~(-1) for 30 days to establish the spermatogenesis impairment model. Normal saline was given to mice in the normal and model control groups. The mice were given the corresponding dose of indole-2,3-dione for 30 days. Sperm, testicular development and fertility were observed. RESULTS Compared with the normal control group, the sperm density and activity rate were reduced in the model control group(P < 0.01).And the numbers of spermatogonia, primary spermatocytes and sperm cells were also reduced(P < 0.01). The sperm density and motility in the indole-2, 3-dione 25, 50, 100 mg·kg~(-1) groups were significantly higher than that in the model control group(P < 0.05 or P < 0.01). The numbers of spermatocytes in the indole-2, 3-dione25 mg·kg~(-1) group were increased(P < 0.05). The numbers of spermatogonia, primary spermatocytes and sperm cells were increased in the indole-2, 3-dione 50, 100 mg·kg~(-1) groups(P < 0.01), and the increase was dose dependent. After paired with females, the fetal mice were(12.7 ± 1.5) in the normal control group. There were1 pregnant mouse and 3 fetal mice in the model control group. The pregnant mice were 2, 3 and 6 and the fetal mice were( 6.1 ± 2.1),( 7.5 ± 1.7) and( 8.6 ± 1.4) in the indole-2, 3-dione 25, 50 and 100 mg·kg~(-1) groups, respectively. CONCLUSION Indole-2, 3-dione can improve the spermatogenic function of the spermatogenesis impairment male mice induced by gossypol acetate.
