根痛平胶囊指纹图谱及5种成分含量测定研究

摘要
目的:建立根痛平胶囊的指纹图谱,同时测定5种成分的含量。方法:采用HPLC法,色谱柱为ACE Execl C_(18)-AR(250 mm×4.6 mm,5μm),柱温:35℃;以乙腈-水为流动相,梯度洗脱,流速:1.0 ml·min~(-1);检测波长:210 nm。利用中药色谱指纹图谱相似度评价系统进行相似度评价。结果:20批样品指纹图谱有18个共有峰,相似度大于0.87。苦杏仁苷、葛根素、芍药苷、甘草苷、川续断皂苷Ⅵ分别在13.22~991.79 ng(r=0.999 8)、21.41~1605.58 ng(r=0.999 8)、52.48~3936.14 ng(r=0.999 9)、4.98~373.70 ng(r=0.999 9)、45.05~3378.39 ng(r=0.999 9)内呈良好的线性关系;平均回收率分别为100.40%(RSD=1.1%),101.90%(RSD=1.3%),100.40%(RSD=1.5%),97.20%(RSD=1.7%),99.09%(RSD=1.9%)(n=6)。结论:指纹图谱特征性强,含量测定方法可靠,可用于根痛平胶囊的质量控制。
Objective:To establish the HPLC fingerprint of Gentongping capsules and determine the contents of 5 chemical compositions.Methods:The HPLC analysis was carried out on a column of ACE Execl C_(18)-AR(250 mm×4.6 mm,5μm)with the column temperature of 35°C.The mobile phase was acetonitrile-water solution with gradient elution at the flow rate of 1.0 ml·min~(-1)and the detection wavelength was 210 nm.The similarity evaluation system for chromatographic fingerprint of TCM was used to calculate the similarity.Results:There were 18 common peaks in the HPLC fingerprints of twenty batches of samples with the similarities more than 0.87.The content of amygdalin,puerarin,paeoniflorin,liquiritin and asperosaponin VI showed a good linear relationship within the range of 13.22-991.79 ng(r=0.999 8),21.41-1 605.58 ng(r=0.999 8),52.48-3 936.14 ng(r=0.999 9),4.98-373.70 ng(r=0.999 9)and 45.05-3 378.39 ng(r=0.999 9),respectively.The average recovery was 100.40%(RSD=1.1%),101.90%(RSD=1.3%),100.40%(RSD=1.5%),97.20%(RSD=1.7%)and 99.09%(RSD=1.9%)(n=6),respectively.Conclusion:The HPLC fingerprints with high characteristics and reliable determination method can be used for the quality control of Gentongping capsules.
