摘要
目的观察补肾固表颗粒对环磷酰胺所致免疫抑制小鼠免疫功能的影响,探讨其作用机制。方法将60只ICR雄性小鼠随机分为空白对照组、模型组、玉屏风组和补肾固表高、中、低剂量组,除空白对照组外,其余各组小鼠腹腔注射环磷酰胺50 mg/(kg2d),连续3 d,建立免疫抑制动物模型,各给药组给予相应药物,连续28 d。末次给药后眼球取血,脱颈处死小鼠后无菌取出脾脏,MTT法检测脾淋巴细胞增殖,LDH法检测NK细胞活力,ELISA检测血清白细胞介素-2(IL-2)和干扰素-γ(IFN-γ)水平。结果与空白对照组比较,模型组小鼠脾淋巴细胞增殖能力、NK细胞活力、IL-2和IFN-γ水平均明显降低(P<0.05,P<0.01);与模型组比较,补肾固表各剂量组小鼠脾淋巴细胞增殖能力、NK细胞活力、IL-2和IFN-γ水平均明显增强(P<0.05,P<0.01,P<0.001),脾脏质量及营养状况亦有改善。结论补肾固表颗粒通过增强免疫低下小鼠脾淋巴细胞转化和杀伤能力,提高血清IL-2和IFN-γ分泌水平,从而改善机体的免疫功能。
        Objective To investigate the effects of Bushen Gubiao Granules on the immune function of immunosuppressed mice induced by cyclophosphamide; To explore the mechanism of immune function regulation.Methods Totally sixty ICR male mice were randomly divided into blank control group, model group, Yupingfeng group, Bushen Gubiao Granules high-, medium-and low-dosage groups. Except for the blank control group, the other groups were injected cyclophosphamide 50 mg/(kg·d) intraperitoneally for 3 consecutive days to establish an animal model of immunosuppression. Each administration group was given the corresponding medicine for gavage for 28 days. After the last administration, the eyeballs were bled; the mice were sacrificed by cervical dislocation; the spleens were aseptically removed. The proliferation of spleen lymphocytes was detected by MTT colorimetry. The killing activity of NK cells was detected by LDH method. Serum levels of IL-2 and IFN-γ were detected by ELISA.Results Compared with the blank control group, the spleen lymphocyte proliferation, NK cell activity, IL-2 and IFN-γlevels in the model group were significantly lower(P<0.05, P<0.01). Compared with the model group, Bushen Gubiao Granules high-, medium-and low-dosage groups could significantly enhance the proliferation of mouse spleen lymphocytes and NK cell killing activity, and increase serum IL-2 and IFN-γ levels(P<0.05, P<0.01, P<0.001). The spleen mass and nutritional status were also improved. Conclusion Bushen Gubiao Granules can enhance thetransformation and killing ability of spleen lymphocytes in immunosuppressed mice, improve the secretion of serum IL-2 and IFN-γ, and promote the immune function of the body.
