摘要
目的建立木鳖子药材皂苷部位超高液相色谱(UPLC)指纹图谱的模式识别分析方法,综合评价其质量。方法采用UPLC-BEH C18色谱柱(2.1 mm×100 mm,1.7μm),流动相为乙腈-0.1%磷酸水溶液,线性梯度洗脱,流速0.25 mL/min,柱温30℃,检测波长203 nm,分别对21批木鳖子样品指纹图谱进行相似度分析、聚类分析和偏最小二乘判别分析(PLS-DA)。结果提取了指纹图谱中的14个共有色谱峰,确认了1个色谱峰;21批木鳖子样品指纹图谱的相似度均大于0.9,其中11批相似度>0.99;聚类分析和PLS-DA可将21批样品分为四大类,PLS-DA色谱峰变量重要性投影(VIP)得分>1.0的有7个。结论本研究建立的模式识别分析方法简便可行,可用于木鳖子药材指纹图谱研究及其质量评价。
        Objective To establish a pattern recognition analysis method to analyze UPLC fingerprints of saponin part in Momordicae Semen; To comprehensively evaluate its quality. Methods The UPLC fingerprints were performed on BEH C18 column(2.1 mm × 100 mm, 1.7 μ m), at 30 ℃. The mobile phases were composed of 0.1% phosphoric acid and acetonitrile, gradient elution. The flow rate was 0.25 mL/min, and the detection wavelength was set at 203 nm. The similarity comparison, hierarchical cluster analysis and partial least squares discriminate analysis(PLS-DA) were applied to study UPLC fingerprints of 21 batches of samples. Results Toally 14 chromatographic peaks were extracted as the common peaks of fingerprints, and one peak was identified. The results showed that similarity degrees of 21 batches of samples were above 0.9, and 11 batches of samples were above 0.99. Cluster analysis and PLS-DA could divide 21 batches of samples into 4 categories, and there were 7 PLS-DA chromatographic peak variable importance projection(VIP) scores >1.0. Conclusion The established pattern recognition analysis method is feasible and simple, and it can be used to evaluate UPLC fingerprints and quality evaluation of Momordicae Semen.
