摘要
目的:探讨膜分离与大孔树脂吸附技术对穿山龙水提液物理化学性质的影响。方法:采取煎煮2次,10倍量水,每次120 min的水煎煮工艺提取穿山龙,分别采用不同型号的大孔树脂(HP-20、HPD-600、D101、AB-8)吸附及膜分离(陶瓷微滤膜0.8μm、0.2μm、0.05μm和中空纤维超滤膜50k Da、6k Da)技术纯化穿山龙水提液,测定其电导率,黏度,浊度等物理化学参数及蛋白质、多糖、总黄酮、薯蓣皂苷等物质的含量。结果:穿山龙水提液经不同技术纯化后黏度无显著影响,电导率和浊度纯化后显著减小;通过各物质保留率可看出D101型大孔树脂适合分离纯化薯蓣皂苷,而HPD-600对于各物质的保留都相对较低,与理化参数结果一致。0.2μm微滤膜更适合主含皂苷水提液的分离纯化。结论:理化参数的改变与大分子物质的含量减小有着明显的对应关系:大分子物质含量降低,电导率与浊度减小。与大孔树脂吸附相比,膜分离技术更适合穿山龙水提液中薯蓣皂苷成分的分离纯化,微滤膜比超滤膜对薯蓣皂苷的保留率更高。
        Objective: To explore the effects of membrane separation technology and macroporous resin adsorption technology on the physical and chemical properties of water extract from Dioscorea nipponica. Methods: Dioscorea nipponica was extracted twice by10-fold amount of water for 120 min each time. The water extract was purified respectively by macroporous resin adsorption( HP-20,HPD-600,D101,AB-8) and membrane separation technology( 0. 8 μm,0. 2 μm and 0. 05 μm ceramic microfiltration membranes,50 k Da and 6 k Da hollow fiber ultrafiltration membranes). The physical and chemical parameters including electrical conductivity,viscosity and turbidity were measured,as well as the contents of proteins,polysaccharides,total flavones and dioscin were detected. Results: The results showed that the electrical conductivity and turbidity decreased greatly,while the viscosity showed no obvious change.The retention of different substances showed that macroporous resin D101 was suitable for the purification of dioscin,while HPD-600 had low retention rate for all the substances,which was consistent with the results of physical and chemical parameters. Microfiltration membrane( 0. 2 μm) was the best choice for the separation and purification of dioscin water extract. Conclusion: The results indicated that the physical and chemical properties of water extract were closely related with the contents of macromolecular substances. The electrical conductivity and turbidity both decrease along with the content decrease of macromolecular substances. Comparing with macroporous resin adsorption technology,membrane separation technology is more suitable for the purification of dioscin in the water extract of Dioscorea nipponica. The microfiltration membrane is better than the ultrafiltration membrane for dioscin reservation.
