摘要
目的比较研究小鼠的雌雄体对有毒有害物质造成的肝损伤的发展过程中血管内皮生长因子(VEGF)的应激反应差别。方法按照体重将昆明小鼠分为3组:正常组、溶剂对照组(腹腔注射实验用橄榄油溶液10μL·g~(-1))、模型组(腹腔注射配制的0. 1%CCl_4橄榄油溶液10μL·g~(-1)),每组雌、雄各3只。在造模24 h后,用试剂盒法测定小鼠血清谷丙转氨酶(GPT);用免疫组化链霉菌抗生物素蛋白-过氧化物酶连结法(SP)法检测小鼠肝中的VEGF的表达量。结果雌性正常组、雌性对照组、雌性模型组、雄性正常组、雄性对照组和雄性模型组血清ALT活性分别为(108±23),(121±36),(1443±40),(106±24),(124±28)和(702±33) U·L~(-1),雌性模型组与雄性模型组比较显著升高,差异有统计学意义(P <0. 01);上述6组的小鼠肝组织中VEGF表达量分别为(12. 5±1. 4),(13. 4±1. 5),(32. 6±2. 9),(13. 2±1. 4),(12. 9±1. 5)和(59. 5±3. 8) cell·mm~(-2),雄性模型组与雌性模型组比较明显升高,差异有统计学意义(P <0. 05)。结论在CCl_4诱导的小鼠化学性肝损伤的过程中,雄性小鼠肝组织中VEGF阳性细胞数量较雌性小鼠表达更为显著,说明雄性小鼠肝损伤后修复能力较强。
        Objective To study the difference in the stress response of vascular endothelial growth factor( VEGF) during the development of liver injury caused by toxic and harmful substances between male and female mice. Methods The Kunming mice were divided into three groups based on their body weight: normal group( natural growth with no treatment),control group( intraperitoneal injection with 10 μL·g~(-1) olive oil solution) and model group( intraperitoneal injection with 10μL·g~(-1)0. 1% CCl_4 olive oil solution). Each group contained 3 males and 3 females. After modeling for 24 hours,the glutamic-pryuvic tronsaminase( GPT) was determined by the kit method. The expression of VEGF in the liver of mice was detected by immunohistochemical streptavidin-peroxidase ligation( SP) method. Results The serum GPT activities in female normal group,female control group,female model group,male normal group,male control group and male model group were( 108 ± 23),( 121 ± 36),( 1443 ± 40),( 106 ± 24),( 124 ± 28) and( 702 ± 33) U·L~(-1) respectively. Comparison between female model group and male model group,the difference was significant( P < 0. 01). The VEGF expression in mouse liver tissue of the above six groups were( 12. 5 ± 1. 4),( 13. 4 ± 1. 5),( 32. 6 ± 2. 9),( 13. 2 ± 1. 4),( 12. 9 ± 1. 5) and( 59. 5 ± 3. 8)cell·mm~(-2). The male model group was significantly higher than female model group( P < 0. 05).Conclusion In the process of chemical liver injury induced by CCl_4 in mice,the expression of VEGF positive cells in liver tissue of male mice was more significant than that of female mice,it indicated that male mice had stronger ability to repair liver injury.
