摘要
目的探讨刀豆蛋白A(Con A)诱导急性免疫性肝损伤小鼠模型的最适剂量及最佳给药途径.方法选用雄性ICR小鼠180只,分为尾静脉注射与腹腔注射两种给药途径,每个给药途径分为6组,对照组、Con A 10,20,30,40和50 mg/kg剂量组,Con A各剂量组注射相应剂量的Con A,对照组注射生理盐水,在注射8 h后,检测各组血清中谷丙转氨酶(ALT)和谷草转氨酶(AST)的水平,采用苏木素-伊红(HE)染色法观察肝组织病理变化.结果与对照组比较,腹腔注射Con A各剂量组的ALT和AST水平无明显变化;尾静脉注射Con A 40 mg/kg剂量组的ALT和AST水平明显升高(P<0. 05);组织学病理显示:肝损伤明显,Con A 10,20,30 mg/kg剂量组ALT和AST水平变化不明显,50 mg/kg剂量组死亡率较高.结论小鼠尾静脉注射40 mg/kg的Con A可成功诱导ICR小鼠的急性肝损伤.
        Objective To investigate the optimal dose and route of Con A administration on acute immunological liver injury in mice. Method 180 ICR mice were divided into intraperitoneal and intravenous injection of two routes of administration. Each route was divided into control group,Con A 10,20,30,40,50 mg/kg dose groups.The mice in Con A group were injected with Con A and those in control group were intraperitoneally injected with normal saline. After the administration for 8 h,the levels of Aspartate aminotransferase(AST) and Alanine aminotransferase(ALT) were measured,the histomorphological liver were observed by hematoxylin and eosin(HE) stain. Results Compared with the control group,there were not significantly changed in the levels of AST and ALT by intraperitoneal injection. While in intravenous injection group,the levels of AST and ALT of mice in Con A 40 mg/kg group were increased obviously and the histomorphological examination of liver indicated that there was an obvious injury,and those in Con A 10,20,30 mg/kg groups were not significantly changed. The mortality rate was higher in 50 mg/kg dose group. Conclusion Intravenous injection of 40 mg/kg Con A can induce acute liver injury in ICR mice.
