厚朴及其提取物体外抗流感病毒H1N1的初步研究

摘要
目的观察厚朴及其提取物体外抗流感病毒H1N1的作用。方法经鸡胚扩增流感病毒H1N1,并测定其感染MDCK细胞的TCID50;采用MTT法检测药物对细胞的毒性及三种给药方式下药物对病毒的抑制作用,并进一步通过流式细胞仪初步探讨药物抗病毒机制。结果H1N1的TCID50为1.3×10~(-9)·L~(-1);厚朴酚、厚朴水浸液及厚朴水煎液的最大无毒浓度分别为1.2μg·mL~(-1)、10.8mg·mL~(-1)及2400μg·mL~(-1);三种给药方式厚朴水煎液对病毒均无作用,厚朴酚有一定的治疗效果,厚朴水浸液有预防和治疗效果;流式细胞仪结果说明厚朴酚和厚朴水浸液的治疗作用可能是通过调整细胞周期、提高细胞增殖指数而实现的。结论对流感病毒H1N1而言,厚朴酚和厚朴水浸液对其有一定的治疗效果,厚朴水浸液对其有一定的预防作用,而厚朴水煎液对其没有作用。
        OBJECTIVE To explore the inhibitory effects of Mangnolia Officinalis and its extractive against influenza virus H1 N1. METHODS Propagation of virus in chick embryo and the value of TCID50 were calculated; The MTT test assay was used to evaluate the toxicity and the inhibitory effect of drugs against H1 N1,and through Flow Cytometry to discuss the antiviral mechanism of Mangnolia Officinalis and its extractive. RESULTS The TCID50 of H1N1 virus to MDCK cells was 1. 3 × 10~(-9)·L~(-1). The TC0 of Magnolol,Mangnolia Officinalis immersion and decoction in MDCK cells were 1. 2μg·mL~(-1),10. 8 mg·mL~(-1) and 2400μg·mL~(-1). Three kinds of way of magnolia officinalis decoction on virus had no effect. Magnolol had a certain therapeutic effect. Mangnolia Officinalis immersion had prevention and therapeutic effect. Flow cytometry results demonstrated the therapeutic effect of Magnolol and Mangnolia Officinalis immersion may be improved through the adjustment of the cell cycle and cell proliferation index. CONCLUSION For H1 N1 influenza virus,Magnolol and Mangnolia Officinalis immersion had certain therapeutic effect,Mangnolia Officinalis immersion had certain preventive effect,and magnolia officinalis decoction has no effect.
