摘要
研究探讨甘露糖结合凝集素(MBL)治疗人工感染绵羊肺炎支原体的效果,揭示MBL蛋白对绵羊支原体肺炎的免疫调节作用,为阐明绵羊肺炎支原体的致病机制提供依据。人工感染绵羊肺炎支原体2周后注射MBL蛋白,对绵羊进行剖检,以及荧光定量PCR检测血浆MBL蛋白和重组MBL蛋白治疗前后,MBL、IFN-γ、TNF-α、IL-2、IL-4、C1、C3的表达变化。结果显示,注射重组MBL蛋白3周后可以减缓患病羊的病程,肺部出血斑和出血点都有轻微的减轻,与无注射治疗的对照组比较,临床症状和病理变化较轻。蛋白治疗7d后血清中IFN-γ、TNF-α、IL-2、IL-4、C3的表达水平升高,14d后呈现下降趋势,但高于对照组。MBL蛋白治疗后,患病绵羊血清中MBL、IL-2、IL-4、IFN-γ、TNF-α、C1和C3表达量均升高,随后减少,证明MBL蛋白能短期内减轻绵羊支原体肺炎症状以及增强机体的细胞和体液免疫,提高机体的抵抗力并改善炎症。
        This research was to investigate the immunomodulatory effects of mannose binding lectin(MBL) on sheep experimentally infected with Mycoplasma ovipneumoniae in the hope of providing support for understanding the pathogenesis of Mycoplasma ovipneumoniae.Before injection of MBL protein,the experimental sheep were artificially infected with Mycoplasma ovipneumoniae,and autopsy was performed on the diseased sheep.Then,qRT-PCR was used to detect changes in IFN-γ,TNF-α,IL-2,IL-4,C1,C3 before and after treatment of Mycoplasma pneumoniae in sheep with plasma MBL protein and recombinant MBL protein.The results showed that three weeks after injection of recombinant MBL protein,the course of the disease in the sheep slowed down,and the pulmonary hemorrhage spots and bleeding spots in the lungs of the experimental sheep were mildly alleviated.Compared with the control group without injection,the clinical symptoms and pathological changes in the sheep were slighter.The expressions of IFN-γ,TNF-α,IL-2,IL-4 and C 3 in their serum increased seven days after protein therapy,but showed a downward trend after the 14 thday of treatment,although the expressions were higher than that of the control group.After treatment with MBL protein,the expression of MBL,IL-2,IL-4,IFN-γ,TNF-α,C 1 and C 3 in the serum of the diseased sheep increased,but then decreased,which implied that the MBL protein alleviated the symptoms of Mycoplasma pneumoniae in sheep in a short time,boosted the cellular and humoral immunity of the animals,enhanced their resistance,and lessened inflammation in their body.
