摘要
目的模拟环境雌激素与冷应激建立复合应激勃起功能障碍(ED)大鼠模型。方法从120只正常雄性大鼠中随机抽取20只为正常组(N组),余100只为建模组(M组)。采用环境雌激素样饮食联合冷应激干预条件建立复合应激大鼠模型,定期观察大鼠生物学体征、行为学变化,并检测应激相关指标,建模24周后,通过阿朴吗啡(APO)勃起和交配实验筛选出ED大鼠模型(ED组),未成ED者为复合应激模型组(S组)。检测各组大鼠血清肾上腺素(E)、去甲肾上腺素(NE)、睪酮(T)、雌二醇(E2)、糖皮质激素(GC)、TNF-α、IL-1β和IL-17α表达水平。结果 (1)M组较N组大鼠体质量增长显著减缓,差异具统计学意义(P<0.05);在中心区活动时间和进入中心区次数显著减少,差异具统计学意义(P <0.05);血清E水平稍有上升,但差异无统计学意义(P>0.05),NE水平显著上升、GC水平显著下降,差异均具统计学意义(P<0.05);阴茎勃起、交配和射精次数显著减少,差异具统计学意义(P<0.05);(2)ED组较N组和S组大鼠血清E2水平稍升高,但差异无统计学意义(P>0.05),T水平显著下降,差异具统计学意义(P <0.05);血清E、NE水平显著上升,GC水平显著下降,差异均具统计学意义(P <0.05);血清TNF-α、IL-1β和IL-17α水平显著上升,差异均具统计学意义(P<0.05)。结论环境雌激素样饮食联合冷应激可成功建立复合应激ED大鼠模型,并为复合应激性ED的基础与应用研究提供了良好的模型。
        Objective To mimic environmental hormones and cold stress to build rats model of erectile dysfunction(ED). Methods 120 normal mature male rats were selected in this study, 20 of them were randomly chosed as the normal group(N group), other 100 as modeling group(M group). The M group rats were treated with cold stress with environment estrogen-like diet, biological characterization and behavioral changes were regularly observed, and stress related indicators were detected until the establishment the stress rats model(after 24 w), among them,we screened ED rats model through the apomorphine(APO) erectile and mating experiments, other rats of them with no ED as complex stress group(S group).Serum epinephrine(E), norepinephrine(NE), testosterone(T), estradiol(E2), glucocorticoids(GC), TNF-α, IL-1β and IL-17α have been detected. Results(1) Compared with N group, rats weight of M group are significantly lower(P < 0.05); activity and times in center significantly reduced(P < 0.05); Serum E level is increased slightly(P > 0.05), NE level increased and the GC level decreased significantly(P < 0.05); Erectile times, mated and ejaculation frequency are significantly reduced(P <0.05).(2) Compared with N and S groups, serum E2 level increased and T levels decreased significantly(P<0.05); E, NE levels increased and GC levels decreased significantly(P<0.05); TNF-α, IL-1β and IL-17 a levels significantly increased(P <0.05). Conclusion Environmental estrogen-like diet combined with cold stress successfully established the ED rats mode, this model provides a good model for the basic and applied research of compound stress ED.
